Sludge bulking has always been a persistent problem in wastewater treatment plants (WWTPs) using activated sludge process due to the lack of understanding its microscopic mechanism. The aim this study was investigate and microbial community behavior full-scale WWTP from perspective quorum sensing, community, metagenomic characteristics. This found that excessive growth Candidatus Microthrix primary case occurred January March. overgrowth also resulted changes diversity structure. Quorum mediated by N-acyl-L-homoserine lactones (AHLs), played role regulating bulking, particularly C6-HSL, while EPS production 3OC8-HSL. In addition, EPS-producing bacteria, AHLs-producing quenching bacteria were detected WWTP. results showed bacterial intercellular communication enhanced control bulking. "Two-component system" responsible for cooperation regulate filamentous growth, it "Oxidative phosphorylation". Meanwhile, "Bacterial secretion system", "Valine, leucine isoleucine biosynthesis", "Lysine as well "Phenylalanine, tyrosine tryptophan biosynthesis" related production.
