Abstract Background Poultry feather waste has a potential for bioenergy production because of its high protein content. This research explored the use chicken hydrolysate methane and hydrogen via anaerobic digestion bioelectrochemical systems, respectively. Solid state fermentation was conducted using recombinant strain Bacillus subtilis DB100 (p5.2). Results In digestion, produced maximally 0.67 Nm 3 CH 4 /kg feathers 0.85 mmol H 2 /day.L concomitant to COD removal 86% 93%, The systems used were microbial fuel electrolysis cells. first cell, electricity with maximum cell 375 mV current 0.52 mA. enhanced rate 7.5 mmol/day.L, density 11.5 A/m power 9.26 W/m . The aprBP gene from Bacillus patagoniensis DB-5, encoding a 378-amino-acid alkaline protease, was cloned and expressed in Escherichia coli . amino acid sequence of APrBP showed 62.8–84.4% identity with the S8 peptidase subtilisin family proteases reported literature. Recombinant purified using Ni-NTA affinity chromatography 45.61% recovery homogeneous band detected at approximately 38 kDa on SDS-PAGE gel. optimum temperature 60°C. presence 2 mM Ca 2+ significantly enhanced optimal thermostability. enzyme demonstrated activity pH 12 maintained high stability 8.0–11.0. Protease stimulated by Mn , Mg Ni TritonX-100, Tween-20 Tween-80, while completely inactivated PMSF, EDTA Cu exhibited good tolerance to oxidizing reducing agents. Notably, protease remarkable 50% (v/v) concentrations several organic solvents, such as methanol, acetone, glycerol, dimethyl sulfoxide, n-hexane, ethyl acetate. efficiently hydrolyzed natural proteins, demonstrating highest catalytic efficiency for casein, excellent hydrolysis bovine serum albumin, hemoglobin, keratin, favorable ability whey proteins. Moreover, molecular docking results revealed stable interactions between proteins keratin. This study indicated that has some useful properties explored its potential bio-additive detergent well utilizing feather waste protein.
