Multiplex biomarker quantitation is ideal for tracking the progression of a disease, but sensitive, specific, and quantitative multiplex biosensing remains challenging. We achieved total internal reflection with two lasers in single planar optical waveguide by coupling 532 nm laser light into diffraction grating 635 directly thin film same waveguide. confirmed that evanescent fields generated can excite fluorescent dyes (Alexa Fluor Alexa 647) held to surface through biotin-streptavidin chemistry. This sensing concept holds significant potential as platform detection.

There remains an unmet need for a fully integrated microfluidic platform that can automatically perform multistep and multireagent immunoassays. Here, we proposed novel online dual-active valve-based centrifugal chip, termed DAVM, automatic point-of-care immunoassay. Practically, the puncture valve, one of dual active valves, is capable achieving precise, on-demand, sequential release prestored reagents, while other valve-reversible valve enables controlled retention drainage reaction solutions. Thereby, our technology mitigates challenges hydrophilic/hydrophobic modifications unstable control performance commonly observed in passive controls. As proof concept, indirect enzymatic immunoblotting technique was employed on DAVM automated immunological analysis eight targets, yielding outcomes within hour.
