Abstract Whole cell microbial biosensors (WCMB) are mostly genetically modified microorganisms used to detect target molecules as indicators of biological and chemical contaminants well in the identification compounds interest food industry. The specificity sensitivity these achieved through design genetic circuits that make use sequences such promoters, terminators, genes encoding regulatory proteins or reporter proteins, among others. Despite advances WCMBs for their application, significant challenges faced, stability, restrictions, need optimize response times so they can be a competitive detection tool market. This review explores technological progress, potential limitations industry, starting by reviewing operating principles biosensors. importance selecting appropriate chassis cells integration recognition elements transducers maximize effectiveness industry is highlighted.

Six novel dichloromethyl-containing polyketides (1-6), including four pyranone analogs (1-4) and two furanone isomers (5, 6), were isolated from Streptomyces agglomeratus 5-2-6, which was obtained the Qinghai-Tibet Plateau, China. Additionally, new linear dichloromethyl compounds (7, 8) via heterologous expression of agg(A-K) in coelicolor M1152. The structures absolute configurations these primarily elucidated high-resolution ESIMS, NMR, Mo2(OAc)4-induced electronic circular dichroism, application modified Mosher's method. Notably, 1-6 feature a moiety, is reported for first time pyranone- furanone-type natural products. Furthermore, results vivo gene deletion, isotope labeling, vitro enzyme reaction experiments revealed rare single-component flavin-dependent halogenase, AggA, catalyzes dichlorination activated sp3-methylene groups.

Short-chain chlorinated aliphatic hydrocarbons (SCAHs), commonly used as industrial reagents and solvents, pose a significant threat to ecosystems human health they infiltrate aquatic environments due extensive usage accidental spills. Whole-cell biosensors have emerged cost-effective, rapid, real-time analytical tools for environmental monitoring remediation. While the broad ligand specificity of transcriptional factors (TFs) often prohibits application such biosensors. Herein, we exploited semirational transition approach in conjunction with positive/negative fluorescence-activated cell sorting (FACS) strategy develop biosensor based on TF AlkS, which is highly specific SCAHs. Furthermore, through promoter-directed evolution, performance was further enhanced. Mutation -10 region constitutive promoter P
