The recognition of small molecules plays a crucial role in disease diagnosis, environmental assessment, and food safety. Currently, their elements predominantly rely on antibodies aptamers while suffering from limitation the complex screening process due to low immunogenicity molecules. Herein, we present top-down computational design strategy for molecule peptides (MRPs) enzyme-peptide self-assembly chemiluminescence biosensing. Taking ochratoxin A (OTA) as an illustrative example, human serum albumin (HSA) was selected parental protein its high affinity OTA binding. Through iterative simulations involving binding domain HSA-OTA complex, our identified specific 15-mer MRP (RLKCASLKFGERAFK), which possesses excellent (38.02 ± 1.24 nM) against OTA. Molecular dynamics revealed that unfolds into flexible short chain with OTA, but exhibits weak or no five structurally similar mycotoxins. Furthermore, developed novel approach mediated by calcium(II) obtain nanoflowers, integrates both element (MRP) signal translator (enzyme) assembled nanoflowers allow MRPs be directly utilized tracer biosensing without labeling secondary antibodies. This computational-to-application offers new route small-molecule recognition.

Melatonin is a key hormone that regulates the sleep-wake cycle and plays an important role in maintaining circadian rhythm sleep onset. The daily rise melatonin secretion associated with increased tendency to sleep, occurring approximately 2 h before bedtime. This correlation between levels onset makes it reliable biomarker for disorders. An accurate assessment of dim light (DLMO) vital understanding timing diagnosing disruptions. However, traditional methods detecting saliva are either complex or lack sensitivity required DLMO, especially at low concentrations. Here, we present novel competitive enzyme-linked aptamer-based assay developed detect saliva. Unlike conventional assays, this technique utilizes chemically synthesized single-stranded DNA RNA aptamers, which bind high specificity sensitivity. measures melatonin, attaining linear dynamic range from 8.62 × 10‒6 M 3.9 10‒11 M, detection limit 2.5 10‒12 (~ 0.57 pg/mL). Additionally, aptamer showed small binding its counter targets acceptable recovery when spiked four times diluted buffer. Overall, portrayed potential aptamers could be beneficial accurately determining particularly individuals very levels, such as elderly those neurodegenerative conditions. Determining precise measurement DLMO will facilitate diagnosis disruption, enabling healthcare providers optimize selection therapeutic behavioural interventions tailored individual's unique rhythm.

Circadian rhythms, essential for regulating human physiology and behavior, are influenced by light exposure, particularly at night. This study examined the impact of red (631 nm) blue (464 LED on melatonin secretion, a key circadian marker. Twelve participants aged 19–55 years were exposed to three hours (9:00 p.m.–midnight), with hourly saliva samples analyzed via ELISA track levels. Initially, levels comparable under both conditions. After one hour, lights suppressed melatonin, but differences emerged after two hours: light-maintained suppression, 7.5 pg/mL, while allowed recovery 26.0 pg/mL (p = 0.019). pattern persisted third hour. Blue had stronger suppression effects, in younger men. These results underscore light’s disruptive effects health highlight as less alternative nighttime environments.

Abstract Beyond storage and transmission of genetic information in cellular life, nucleic acids can perform diverse interesting functions, including specific target recognition biochemical reaction acceleration; the versatile biopolymers, however, are acutely vulnerable to hydrolysis-driven degradation. Here, we demonstrate that cage effect choline dihydrogen phosphate permits active folding like water, but prevents their phosphodiester hydrolysis unlike water. The choline-based ionic liquid not only serves as a universal inhibitor nucleases, exceptionally extending half-lives up 6 500 000 times, highly useful tasks (e.g. mRNA detection molecular beacons, ligand aptamers, transesterification ribozymes) be also conducted with well-conserved affinities specificities. As liberated from function loss degradation risk, presence undesired unknown nucleases does undermine desired functions without artifacts even nuclease cocktails human saliva.
