Tertiary lymphoid structures (TLSs) are defined as aggregates formed in non-hematopoietic organs under pathological conditions. Similar to secondary (SLOs), the formation of TLSs relies on interaction between tissue inducer (LTi) cells and organizer (LTo) cells, involving multiple cytokines. Heterogeneity is a distinguishing feature TLSs, which may lead differences their functions. Growing evidence suggests that associated with various diseases, such cancers, autoimmune transplant rejection, chronic inflammation, infection, even ageing. However, detailed mechanisms behind these clinical associations not yet fully understood. The by TLS maturation localization affect immune function also unclear. Therefore, it necessary enhance understanding development at cellular molecular level, allow us utilize them improve microenvironment. Abstract N 1 -methyladenosine (m A) RNA methylation is critical for regulating mRNA translation; however, its role in the development, progression, and immunotherapy response of head neck squamous cell carcinoma (HNSCC) remains largely unknown. Using Tgfbr1 Pten conditional knockout (2cKO) mice, we found neoplastic transformation oral mucosa was accompanied by increased m A modification levels. Analysis A-associated genes identified TRMT61A as a key writer linked to cancer progression poor prognosis. Mechanistically, TRMT61A-mediated tRNA-m promotes MYC protein synthesis, upregulating programmed death-ligand (PD-L1) expression. Moreover, levels were also elevated tumors treated with oncolytic herpes simplex virus (oHSV), contributing reactive PD-L1 upregulation. Therapeutic inhibition sustained oHSV-induced antitumor immunity reduced tumor growth, representing promising strategy alleviate resistance. These findings indicate that can prevent immune escape after oHSV therapy reducing expression, providing mutually reinforcing combination approach.

Abstract Covalent organic frameworks (COFs) exhibit immense potential for phototherapy due to their exceptional light absorption, photostability, and biocompatibility. Nevertheless, tumor cells can develop resistance after an initial response, posing a significant challenge complete eradication. In this study, COF‐based photosensitizer, TCN‐PPDA‐COF, is designed with outstanding photothermal properties delivered tumors using microneedles (MN) enhance cancer phototherapy.
