Neoadjuvant chemotherapy (NACT) is the standard-of-care treatment for patients with locally advanced breast cancer (LABC), providing crucial benefits in tumor downstaging. Clinical parameters, such as molecular subtypes, influence therapeutic impact of NACT. Moreover, severe adverse events delay process and reduce effectiveness therapy. One hundred paired blood samples were collected from 50 LABC before after a complete NACT cycle. Untargeted metabolomics was used by liquid chromatography-mass spectrometry (LC–MS) to investigate relationship between dynamically changing metabolites serum toxicities Firstly, we observed significant alterations metabolite levels pre- post-NACT, predominant enrichment sphingolipid amino acid metabolism pathways. Second, pre-treatment successfully predicted response hematotoxicities In particular, subtype variations favorable linked acyl carnitine levels. Finally, discovered that effects could be attributed essential metabolism. This study elucidated dynamic treatment, possibility developing responsive signatures personalized treatment.

The integration of advanced analytical platforms, such LC–MS GC–MS, coupled with metabolomics, propelled the qualitative quantitative characterization TCM’s complex components. This review comprehensively examines applications MS-based elucidating TCM efficacy, spanning chemical composition analysis, molecular target identification, mechanism-of-action studies, syndrome differentiation.
