Pathogenic bacterial infections pose a significant threat to human life, health, and socioeconomic development, with those arising from Escherichia coli (E. coli) O157:H7 being particularly concerning. Herein, customized aggregation-induced emission luminogens (AIEgen)-based signaling tags (TPA-galactose) were combined microfluidic chip for the determination of E. O157:H7. TPA-galactose undergoes hydrolysis by β-galactosidase, resulting in formation highly fluorescent TPA–OH AIE characteristics. Phages covalently bound surface magnetic beads specifically capture lyse O157:H7, releasing endogenous fluorescence intensity facilitates The process achieves sensitivity 45 CFU/mL min, requiring no DNA extraction or amplification, utilizing minimal sample volume, enabling accurate one-stop quantification live This strategy enables rapid on-site environmental, food, clinical samples, significantly enhancing public health safety.

Abstract Bacterial infection is a crucial factor resulting in public health issues worldwide, often triggering epidemics and even fatalities. The accurate, rapid, convenient detection of viable bacteria an effective method for reducing infections illness outbreaks. Here, unsupervised learning–assisted surface acoustic wave–interdigital transducer‐driven nano‐lens holography biosensing platform developed the ultrasensitive amplification‐free bacteria. monitoring device integrated with effect can achieve holographic imaging polystyrene microsphere probes ultra‐wide field view (∽28.28 mm 2 ), sensitivity limit as low 99 nm. A lightweight learning hologram processing algorithm considerably reduces training time computing hardware requirements, without requiring datasets manual labels. By combining phage–mediated bacterial DNA extraction enhanced CRISPR–Cas12a systems, this strategy successfully achieves Salmonella various real samples, demonstrating accuracy validated qPCR benchmark method. This approach has cost (∽$500) rapid (∽1 h) highly sensitive (∽38 CFU mL −1 allowing emerging powerful tool food safety inspection clinical diagnosis.
