Abstract The emerging monkeypox virus (MPXV) has raised global health concern, thereby highlighting the need for rapid, sensitive, and easy-to-use diagnostics. Here, we develop a single-step CRISPR-based diagnostic platform, termed SCOPE (Streamlined CRISPR On Pod Evaluation platform), field-deployable ultrasensitive detection of MPXV in resource-limited settings. viral nucleic acids are rapidly released from rash fluid swab, oral saliva, urine samples 2 min via streamlined lysis protocol, followed by 10-min recombinase polymerase amplification (RPA)-CRISPR/Cas13a reaction. Abstract The World Health Organization’s designation of mpox as a public health emergency international concern in August 2024 underscores the urgent need for effective diagnostic solutions to combat this escalating threat. rapid global spread clade II mpox, coupled with sustained human-to-human transmission more virulent I Democratic Republic Congo, highlights critical gap point-of-care diagnostics emergent disease. Abstract Genomic data collected from viral outbreaks can be exploited to reconstruct the dispersal history of lineages in a two-dimensional space using continuous phylogeographic inference. JYNNEOS, a third-generation smallpox vaccine, is integral to monkeypox virus (MPXV) control efforts, but the durability of this modified vaccinia Ankara-Bavarian Nordic (MVA-BN) vaccine's effectiveness undefined. We optimized and used plaque reduction neutralization test (PRNT) with authentic clade IIa MPXV assess antibody responses over 12 months in 8 donors vaccinated 2 doses JYNNEOS. One donor previously received ACAM2000 vaccine; 7 were vaccine-naive. IgG (L1, B5, A33) or (E8, H3, A35) antigens PRNT titers both viruses peaked at weeks postvaccination waned rapidly thereafter naive donors. especially low; no demonstrated 90% reduction. Abstract The 2022 global mpox epidemic was caused by transmission of MPXV clade IIb, lineage B.1 through sexual contact networks, with New York City (NYC) experiencing the first and largest outbreak in United States. Abstract Background In May 2022, mpox (formerly monkeypox) began spreading globally through LGBTQ+ sexual networks. By August New York City (NYC) became the global epicenter of outbreak, with highest number cases reported in United States. Here, we quantify vaccination effort, focusing on flexible and community-responsive mobile vaccination. Methods We describe an on-site strategy at commercial sex venues, nightlife pride health centers, during 1–November 15, 2022. Data were collected doses, demographics, event size to determine evaluate vaccine uptake. Results The resulted 3358 JYNNEOS doses administered 363 events 58 locations, including 22 2 venues.
