Abstract Reprogramming of lipid metabolism is a newly recognized hallmark malignancy. Increased uptake, storage and lipogenesis occur in variety cancers contribute to rapid tumor growth. Lipids constitute the basic structure membranes also function as signaling molecules energy sources. Sterol regulatory element‐binding proteins (SREBPs), family membrane‐bound transcription factors endoplasmic reticulum, play central role regulation metabolism. Recent studies have revealed that SREBPs are highly up‐regulated various promote SREBP cleavage‐activating protein key transporter trafficking activation well critical glucose sensor, thus linking de novo synthesis. Targeting altered metabolic pathways has become promising anti‐cancer strategy. This review summarizes recent progress our understanding malignancy, highlights potential molecular targets their inhibitors for cancer treatment.

The integration of mass spectrometry-based proteomics with next-generation DNA and RNA sequencing profiles tumors more comprehensively. Here this "proteogenomics" approach was applied to 122 treatment-naive primary breast cancers accrued preserve post-translational modifications, including protein phosphorylation acetylation. Proteogenomics challenged standard cancer diagnoses, provided detailed analysis the ERBB2 amplicon, defined tumor subsets that could benefit from immune checkpoint therapy, allowed accurate assessment Rb status for prediction CDK4/6 inhibitor responsiveness. Phosphoproteomics uncovered novel associations between suppressor loss targetable kinases. Acetylproteome highlighted acetylation on key nuclear proteins involved in damage response revealed cross-talk cytoplasmic mitochondrial metabolism. Our results underscore potential proteogenomics clinical investigation through annotation pathways biological features remarkably heterogeneous malignancy.

Abstract Cuproptosis was a copper-dependent and unique kind of cell death that separate from existing other forms death. The last decade has witnessed considerable increase in investigations programmed death, whether copper induced an independent form long been argued until mechanism cuproptosis revealed. After that, increasing number researchers attempted to identify the relationship between process cancer. Thus, this review, we systematically detailed systemic cellular metabolic processes copper-related tumor signaling pathways. Moreover, not only focus on discovery its mechanism, but also outline association cancers. Finally, further highlight possible therapeutic direction employing ion ionophores with cuproptosis-inducing functions combination small molecule drugs for targeted therapy treat specific

Abstract The growing field of immunometabolism has taught us how metabolic cellular reactions and processes not only provide a means to generate ATP biosynthetic precursors, but are also way controlling immunity inflammation. Metabolic reprogramming immune cells is essential for both inflammatory as well anti-inflammatory responses. Four therapies, DMF, Metformin, Methotrexate Rapamycin all work by affecting metabolism and/or regulating or mimicking endogenous metabolites with effects. Evidence emerging the targeting specific events strategy limit inflammation in different contexts. Here we discuss these recent developments speculate on prospect effort develop novel therapeutics. As accumulating evidence roles an intricate elaborate network processes, including lipid, amino acid nucleotide provides key focal points developing new here turn our attention glycolysis TCA cycle examples intermediates enzymes can potential therapeutic targets.
