Abstract Aims The association between the gut microbiota and non-alcoholic fatty liver disease (NAFLD) has been documented; however, causal relationship them remains unclear. aim of this study was to explore NAFLD using Mendelian randomization. Methods We conducted a randomization study, data (n = 18340) from MiBioGen consortium as exposure dataset FinnGen R9 release 377,277) outcome. Inverse variance weighted employed primary analysis method, sensitivity performed. Additionally, network graph resembling phylogenetic tree created further reveal evolutionary relationships among microbiota. Results Six bacterial features exhibited with NAFLD: Actinomycetales (OR 1.50, 95% CI: 1.01–2.21, p 0.043), Actinomycetaceae 1.49, 1.01–2.20, Actinomyces 1.36, 1.09–1.71, 0.006), Prevotella 7 1.19, 1.01–1.39, 0.039) potential detrimental effects on NAFLD, whereas Anaerofilum 0.85, 0.72–0.99, 0.042) Gordonibacter 0.84, 0.72–0.98, 0.024) protective effects.
