Sortase A (SrtA) of Staphylococcus aureus has long been shown to be a relevant molecular target for antibacterial development. Moreover, the designed SrtA inhibitors act via antivirulence mechanism, potentially causing less evolutional pressure and reduced antimicrobial resistance. However, no marketed drugs or even drug candidates have reported until recently, despite numerous efforts in field. tough rational structure-based design (SBDD), which hampers regular development small-molecule using available arsenal discovery tools. Recently, several oligopeptides resembling sorting sequence LPxTG (Leu-Pro-Any-Thr-Gly) native substrates were active micromolar range. Despite good experimental those works, their modeling parts are still not convincing enough used as basis modification peptidic inhibitors. In this work, we propose use ensemble docking approach, conformations extracted from dynamics simulation LPRDA (Leu-Pro-Arg-Asp-Ala)-SrtA complex, effectively represent most significant diverse conformations. The developed protocol is describe known data well then applied series new peptidomimetic molecules oligopeptide structures previously order prioritize work further synthesis activity testing. proposed approach compared existing alternatives, directions its outlined.

Background Urinary tract infections (UTIs) represent one of the most prevalent bacterial infections, with Enterococcus species now recognized as second leading cause these infections. This study focused on symptomatic UTI cases to investigate risk factors associated faecalis clinical isolates in patients from Tehran, Iran. Methods Urine samples were collected presenting UTIs. The identification E. was performed using standard microbiological techniques, confirmation via polymerase chain reaction (PCR). Antibiotic susceptibility testing conducted Kirby–Bauer disc diffusion method. presence virulence genes determined through PCR, and biofilm formation assessed microtiter plate Additionally, multi-locus sequence typing (MLST) utilized genotype linezolid-resistant isolates. Results Out 300 , identified causative agent 160 instances. Notably, a high proportion exhibited resistance tetracycline (83.8%) minocycline (82.5%). Linezolid observed 1.3% ( n = 2) Conversely, highest rates for vancomycin, penicillin G, ampicillin, nitrofurantoin, each demonstrating 98.8% rate. Biofilm detected 25% A significant majority (93.8%) harbored efbA ace genes, varying frequencies esp (72.5%), asa1 (61.2%), cylA (52.5%), gelE (88.8%) genes. MLST analysis demonstrated that both isolates, characterized by strong assigned ST150 lineage, which has not been previously documented settings. Conclusion emergence clonal underscores its significance, particularly relation linezolid . adds growing body evidence linking specific lineages antibiotic resistance, highlighting critical need ongoing surveillance molecular characterization resistant pathogens.
