Paramignya trimera ( P. (Oliv.) Guillaum), a Vietnamese plant has showed potential anticancer activities. Extracts from the plant's roots, stems, and leaves have shown cytotoxicity against variety of cancer cell lines. The mechanisms behind these actions include triggering apoptosis, blocking invasion, targeting particular signaling pathways. This review collected data available literature on trimera's properties compiled database its isolated compounds. Results indicate that extracts specific compounds, such as coumarins acridones, possess potential. Particularly, acridones exhibited comparable or superior HepG2 MCF-7 cells (IC 50 values 0.43-1.19 μM 0.22-0.40 μM, respectively) compared to doxorubicin 0.28 1.31 μM). Coumarins like 8-geranylumbelliferone ostruthin target IKKβ, while paratrimerin W shows various cancers. Paratrimerin I exhibit due N-methyl, C-4 methoxy, C-5 hydroxy groups in acridone skeleton. However, further research is needed elucidate precise action evaluate safety efficacy trimera-derived compounds clinical settings. identification 146 provides valuable resource for future drug discovery efforts.

Background/Objectives: Metabolic-dysfunction-associated steatotic liver disease (MASLD) progresses from hepatic steatosis to hepatocellular carcinoma (HCC) as a result of systemic immunometabolic dysfunction. This review summarizes the key roles innate and adaptive immune mechanisms driving injury, fibrogenesis, carcinogenesis in MASLD. Methods: A comprehensive literature was performed using PubMed identify relevant published studies. Eligible articles included original research clinical studies addressing immunological metabolic MASLD, well emerging therapeutic strategies. Results: We highlight cytokine networks, gut–liver axis, cell reprogramming. Emerging strategies, including inhibitors, anti-fibrotic agents, modulators, nutraceuticals, offer several indications for attenuating MASLD progression reducing prevalence extrahepatic manifestations. Conclusions: Given heterogeneity personalized combination-based approaches targeting both inflammation stress are essential effective management prevention complications.

Non-alcoholic fatty liver disease (NAFLD), characterized by the excessive accumulation of fat within cytoplasm hepatocytes (exceeding 5% weight) in individuals without significant alcohol consumption, has rapidly evolved into a pressing global health issue, affecting approximately 25% world population. This condition, closely associated with obesity, type 2 diabetes, and metabolic syndrome, encompasses spectrum disorders ranging from simple steatosis inflammation to non-alcoholic steatohepatitis (NASH) cirrhotic disease. Recent research illuminated complex interplay between immune responses pathogenesis NASH, underscoring critical role played T B lymphocytes. These cells not only contribute necroinflammatory changes hepatic lobules but may also drive onset progression fibrosis. narrative review aims provide comprehensive exploration effector mechanisms employed cells, their respective subpopulations NASH. Understanding immunological complexity NASH holds profound implications for development targeted immunotherapeutic strategies combat this increasingly prevalent burdensome

Metabolic dysfunction-associated steatotic liver disease (MASLD) currently represents the predominant cause of chronic and is closely linked to a significant increase in risk hepatocellular carcinoma (HCC), even absence cirrhosis. In this retrospective multicenter study, machine learning (ML) methods were employed investigate relationship between metabolic profile prognosis at diagnosis total 219 HCC patients. The eXtreme Gradient Boosting (XGB) method demonstrated superiority identifying mortality predictors our Etiology was most determining prognostic factor followed by Barcelona Clinic Liver Cancer (BCLC) Eastern Cooperative Oncology Group (ECOG) classifications. Variables related development hepatic steatosis syndrome, such as elevated levels alkaline phosphatase (ALP), uric acid, obesity, alcohol consumption, high blood pressure (HBP), had impact on prediction. This study underscores importance syndrome progression secondary MASLD. use ML techniques provides an effective tool improve stratification individualized therapeutic management these

Abstract The liver routinely encounters antigens from the gut, triggering pro- inflammatory responses. Unresolved inflammation can lead to damage, steatosis, fibrosis, cirrhosis, and eventually hepatocellular carcinoma (HCC). HCC is influenced by various immune cells, including invariant natural killer T (iNKT) which exhibit both innate adaptive immunity traits. Here, we examined iNKT cell dynamics in a diethyl-nitrosamine (DEN)-induced mouse model. We observed significant reduction numbers livers due apoptosis impaired cytokine production. CD1d -deficient mice, lack displayed delayed tumor initiation lower foci number. However, these tumors were larger size characterized enhanced proliferation immunosuppression. Interestingly, adoptive transfer of healthy cells post-tumor establishment reduced burden, highlighting their potential therapeutic role. Our findings suggest that contribute early development, while later stages they help control growth, thus underscoring complex role carcinogenesis. Further understanding functions may inform novel immunotherapeutic strategies for management.
