Benzophenone-3 (BP-3), commonly used as a UV filter in personal care products and stabilizer, is an alleged endocrine disruptor with potential neurodevelopmental impacts. Despite its abundance the environment, studies on effect brain development are scarce, especially terms of multigenerational impact. In this work, for first time, we examined neurotoxic pro-apoptotic effects BP-3 mouse regions (cerebral cortex hippocampus) both (F1) second (F2) generations after maternal exposure to environmentally relevant levels. Exposure to endocrine disrupting chemicals (EDCs), particularly benzophenone (BP)-type UV filters, has been epidemiologically linked endometrium-related reproductive risks in women. However, their effects on hormone-driven endometrial events and key receptor signaling at the human cellular level remain unexplored. Herein, using primary stromal cells (HESCs), we investigated of five BP congeners deciphered underlying mechanism decidualization, a functional change endometrium preparing for pregnancy. BP-8, its two metabolites, BP-3, BP-1 10 nM significantly disrupted progesterone-dependent decidualization HESCs, marked by 1.5–1.8-fold 2.2–2.6-fold upregulation IGFBP-1 LEFTY, respectively. Decidual transcriptional activators, WNT-FOXO1, were induced BPs, which are implicated G2 phase cell arrest (from 3.26% 8.93%) apoptosis 12.29% 25.61%). Mechanistically, inhibition estrogen α (ERα) effectively alleviated these decidual effects. BPs increased transcription ERα progesterone (PR) enhanced nuclear translocation interaction between PR during decidualization. The ERα-mediated enhancement activity was further validated response element-luciferase transfected cells. Collectively, our findings elucidate molecular pathway through disrupt via ERα/PR/FOXO1, providing critical mechanistic insights risk assessment structurally related EDCs.
