Cancer
treatments,
including
cytotoxic
therapy,
often
result
in
male
infertility,
necessitating
the
development
of
safe
and
effective
strategies
to
preserve
reproductive
potential
during
chemotherapy.
Notably,
our
study
uncovers
repurposing
riluzole,
an
FDA-approved
drug
for
amyotrophic
lateral
sclerosis
(ALS),
enhancing
spermatogenesis.
Hence,
this
research
aims
explore
feasibility
utilizing
riluzole
alleviate
infertility
induced
by
busulfan
(BSF),
a
commonly
used
chemotherapy
drug.
We
established
BSF-induced
oligospermia
model
4-week-old
mice
found
that
could
effectively
counter
detrimental
effects
BSF
on
sperm
production
with
oligospermia.
By
restoring
blood-testis
barrier
(BTB)
functionality,
improves
quality
reduces
testicular
atrophy.
Through
transcriptomic
molecular
docking
analyses,
we
identify
transient
receptor
canonical
subfamily
member
5
(TRPC5)
as
target
riluzole-mediated
regulation
function.
These
findings
propose
promising
therapeutic
option
chemotherapy-induced
thereby
addressing
fertility
challenges
associated
cancer
treatments.
