Cancer treatments, including cytotoxic therapy, often result in male infertility, necessitating the development of safe and effective strategies to preserve reproductive potential during chemotherapy. Notably, our study uncovers repurposing riluzole, an FDA-approved drug for amyotrophic lateral sclerosis (ALS), enhancing spermatogenesis. Hence, this research aims explore feasibility utilizing riluzole alleviate infertility induced by busulfan (BSF), a commonly used chemotherapy drug. We established BSF-induced oligospermia model 4-week-old mice found that could effectively counter detrimental effects BSF on sperm production with oligospermia. By restoring blood-testis barrier (BTB) functionality, improves quality reduces testicular atrophy. Through transcriptomic molecular docking analyses, we identify transient receptor canonical subfamily member 5 (TRPC5) as target riluzole-mediated regulation function. These findings propose promising therapeutic option chemotherapy-induced thereby addressing fertility challenges associated cancer treatments. Moreover, streamline process, providing cost-effective approach reduced risk compared developing entirely new drugs.

Abstract STUDY QUESTION Is ultra-processed food (UPF) consumption associated with semen quality parameters? SUMMARY ANSWER Higher UPF was inversely total sperm count, concentration, and motility in men of reproductive age. WHAT IS KNOWN ALREADY The UPF, which has been rising during the last decades, demonstrated to be positively several chronic diseases such as diabetes or cardiovascular diseases. However, scientific evidence on its potential impact remains notably limited. This federal state covers urban rural compartments including mountainous river areas. A temporal spatial analysis of internal relationships was performed using (a) cross-correlations, both grand ensemble as well within subsets, (b) Local Indications Spatial Association (LISA) method. Meteorological strongly correlated themselves time space. We found strong interaction between ozone, with correlation coefficients varying over time. The ranged from negative correlations January (−0.84), April (−0.47), October (−0.54) positive July (0.45). cross-correlation plot showed noticeable change direction . Bisphenol A (BPA) is a widespread industrial chemical, used as the key monomer of polycarbonate plastics and epoxy resins. BPA has been detected in human seminal fluid correlated with changes sperm parameters, crucial determining male fertility. In this study, semen samples were collected from 100 patients aged 29-47 years undergoing fertility assessment between 2021 2023 analyzed according to WHO guidelines. levels plasma then measured through an enzyme-linked immunosorbent assay (ELISA) compared quality metrics. The relative mRNA/miRNA expression genes associated reproduction, including androgen receptor, miR-34c, miR-21, miR-130a, was quantified groups high or low content. Our results revealed that increased age negatively counts (p<0.05). negative correlation remained significant when age-matched. No other relationships motility, morphology DNA fragmentation observed. qPCR analysis showed receptor mRNA significantly greater Moreover, we found miR-21 miR-130a also upregulated higher group These display relationship content provide insights into molecular mechanisms which may be affecting reproductive capability. Ultimately, research can potentially drive guidelines exposure limits for exposure.
