Abstract Oral squamous cell carcinoma (OSCC) is the most common manifestation of oral cancer. It has been proposed that periodontal pathogens contribute to OSCC progression, mainly by their virulence factors. However, main pathogen and its mechanism modulate cells remains not fully understood. In this study we investigate host-pathogen pathways in computational proteomics behind cancer progression microbiome. The were analyzed secretome biopsies from patients with healthy controls mass spectrometry. Then, functional assays performed evaluate highlighted Host proteins associated LPS response, migration/adhesion, metabolism amino acids significantly upregulated human proteome, whereas complement cascade was downregulated malignant samples. microbiome analysis revealed large number variety peptides Fusobacterium nucleatum ( F. ) samples, which several enzymes L-glutamate degradation pathway found, indicating used as an energy source, catabolized into butyrate bacteria. fact, observed modulates cystine/glutamate antiporter line increasing SLC7A11 expression, promoting efflux favoring bacterial infection. Finally, our results showed metabolic derivates promote tumor spheroids growth, spheroids-derived detachment, epithelial-mesenchymal transition Galectin-9 upregulation. Altogether, promotes pro-tumoral

Abstract The present study explored the anticancer activity of a Chitosan-based nanogel incorporating thiocolchicoside and lauric acid (CTL) against oral cancer cell lines (KB-1). Cell viability, AO/EtBr dual staining cycle analysis were done to evaluate impact CTL on cells. Real-time PCR was performed analyze proapoptotic antiapoptotic gene expression in CTL-treated KB-1 Further, molecular docking conducted explore interaction our key ingredient, its binding affinities. demonstrated potent by inhibiting proliferation inducing arrest Gene indicated alterations Bax Bcl-2 genes; treatment increased mRNA inhibited expression, which showed potential mechanisms nanogel's action. It found that can stabilize protein's function or restore it as tumour suppressor. exhibited excellent cytotoxicity effects, making candidate for non-toxic chemotherapy nanomedicine. Furthermore, ability modulate highlights targeted therapy. This research contributes growing interest nanogels their applications treatment.

Oral squamous cell carcinoma (OSCC) is one of the most common malignancies head and neck (HNSCC). HNSCC recognized as eighth commonly occurring cancer globally in men. It essential to distinguish between cancers arising regions due significant differences their etiologies, treatment approaches, prognoses. As Cancer Genome Atlas (TCGA) dataset available HNSCC, survival analysis prognosis OSCC patients based on TCGA for discovering gene expression-based prognostic biomarkers limited. To address this paucity, we aimed provide comprehensive evidence by recruiting studies that have reported new biomarkers/signatures establish a model predict patients. Using PubMed search, identified 34 been using least absolute shrinkage selection operator (LASSO)-based Cox regression analyses signature related different pathways from past 4 years. Our review was focused summarizing these signatures implications targeted therapy FDA-approved drugs. Furthermore, conducted an LASSO signatures. findings revealed 13 correlated greater number regulatory T cells (Tregs) protective with increased recurrence-free overall rates. Conversely, two displayed opposing trend cases OSCC. We will also explore how dysregulation impacts immune status, promoting tumor evasion or, conversely, enhancing surveillance. Overall, insight future anti-cancer therapies potential associated poor

Currently, cell therapeutics in oral cancer less efficient due several limitations, follows: (a) lower percentages peripheral blood cells; (b) limited survival decreased especially tumor microenvironment; (c) a lack tools or methodologies expand activate levels that are required for effective targeting cancer. To overcome these we established demonstrated novel technology activating expanding highly functional coined supercharged (sNK) cells.
