Abstract As an interspecies signalling molecule, indole that is also regulating the microbial community quorum sensing (QS) can be indispensable factor to influence performance of anaerobic digestion process. Mainly released by Gram-negative bacteria, impact regulation towards methane production in such system hardly exposed. This research intends analyse affected fluctuations waste sewage sludge (WSS) response QS repression and its analogs: 4-fluoroindole (4-FI), 5-fluoroindole (5-FI), 6-fluoroindole (6-FI), 3- indoleacetic acid (3-IAA), indoleacrylic (3-IARA). Illumina MiSeq platform was utilized delve into active microbes, with ribonucleic (RNA) used as template generate 16S metagenomic library. results, all them inhibit generation even substrates (acetic acid) availability, this phenomenon led slight imbalance Gram-positive/negative bacterial composition inactivation viable core fermenters: Firmicutes, Proteobacteria Chloroflexi. Plus, following compounds, indole, 3-IAA, 6-FI directly disrupted productions both acetoclastic hydrogenotropic methanogenic archaea. Methanosarcina acetivorans C2A a prominent synthesizer characterised terms viability synthesis activity against them. Notably, non-viable presence thus lowering production. From here, affirmation on direct indirect inhibitions archaea derivatives will pave way for valuable future exploration mechanism mapping during methanogenesis.

Anaerobic digestion is a crucial process for treating organic waste, such as wastewater sludge, agricultural residues and food waste. While the influence of physicochemical parameters on prokaryotic community composition in anaerobic digesters has been extensively characterized, role biotic interactions shaping communities remains poorly understood. This study addresses this knowledge gap by analyzing complete active microbiome nine full-scale digesters. Our findings reveal that eukaryotes, consisting primarily protists fungi, account approximately 40 % RNA sequence reads alongside dominant Archaea, indicating their substantial process. results suggest chosen sludge retention time during indirectly affects archaeal thus treatment efficacy cascading through highlighting integral system. highlights critical eukaryotes regulating indirect contribution to optimization efficiency.
