Abstract Background Genetic and environmental factors are implicated in many developmental processes. Recent evidence, however, has suggested that epigenetic changes may also influence the onset of puberty or susceptibility to a wide range diseases later life. The present study aims investigate genomic DNA methylation profiles associated with pubertal analyzing human peripheral blood leukocytes from three different groups subjects: 19 girls central precocious (CPP), 14 healthy prepubertal matched by age 13 stage. For this purpose, comparisons were performed between pre- controls identify normal transition CPP versus controls. Results Analysis identified 1006 differentially methylated CpG sites, 86% them found be hypermethylated Some these sites reside genes menarche transcription involved process development. methylome patients showed 65% 55% hypomethylated compared controls, respectively. In addition, interestingly, our results revealed presence 43 coding for zinc finger (ZNF) proteins. Gene ontology IPA analysis studied significant enrichment some pathways related neuronal communication (semaphorin gustation pathways), estrogens action, cancers (particularly breast ovarian) metabolism sirtuin). Conclusions indicate regulation humans is specific changes. Puberty is a key phase of growth and development, characterized by psychophysical transformations. It driven combination genetic, hormonal, environmental variables. Epigenetic mechanisms, including histone post-translational modifications chromatin remodeling, microRNAs, DNA methylation, play important roles in orchestrating the developmental processes. We describe factors that may interact with genetics, influencing puberty onset, focusing particular on epigenetic mechanisms can help understand timing variations lead to precocious or delayed puberty.

Background: Female reproductive aging remains irreversible. More evidence is needed on how polyunsaturated fatty acids (PUFAs) affect the female lifespan. Objectives: To identify and validate specific PUFAs that can influence timing of menarche menopause in women. Methods: We utilized a two-sample Mendelian randomization (MR) framework to evaluate causal relationships between various longevity, defined by age at (AAM) natural (ANM). Our analyses leveraged summary statistics from four genome-wide association studies (GWASs) plasma concentrations 10 PUFAs, including 8866 121,633 European individuals 1361 East Asian individuals. Large-scale GWASs for traits provided genetic data AAM ANM over 202,323 females 43,861 females. Causal effects were estimated beta coefficients, representing, each increase standard deviation (SD) PUFA concentration, yearly or ANM. Replications, meta-analyses, cross-ancestry assessed inference. Conclusions: Higher DHA was identified be associated with delayed without affecting menarche, offering potential intervention target extending longevity.
