This review delves into the detrimental impact of alcohol consumption on internal organs and reproductive health, elucidating underlying mechanisms involving Toll-like receptor 4 (TLR4)/Nuclear factor kappa light chain enhancer activated B cells (NF-kB) pathway Cytochrome P450 2E1 (CYP2E1)/reactive oxygen species (ROS)/nuclear erythroid 2-related 2 (Nrf2) pathways. The TLR4/NF-kB pathway, crucial for inflammatory immune responses, triggers production pro-inflammatory agents type-1 interferon, disrupting balance between antioxidant responses when tissues are chronically exposed to alcohol. Alcohol-induced dysbiosis in gut microbes heightens wall permeability pathogen-associated molecular patterns (PAMPs), leading liver cell infection subsequent inflammation. Concurrently, CYP2E1-mediated metabolism generates ROS, causing oxidative stress damaging cells, lipids, proteins, deoxyribonucleic acid (DNA). To counteract this imbalance, Nrf2 regulates gene expression, inhibiting progression promoting responses. Excessive intake results elevated enzymes (ADH, CYP2E1, catalase), NADH, acetaldehyde, acetate, damage vital such as heart, brain, lungs.
