Perioperative neurocognitive disorder (PND) is the main cause of poor postoperative recovery in elderly patients with age-related reductions androgen levels. However, underlying mechanisms have not been completely elucidated.A mouse model PND was constructed using abdominal surgery. Dihydrotestosterone (DHT), as primary androgen, can improve cognitive function mice PNDs by reducing REDOX damage. To clarify role circular RNA (circRNA) DHT improving PND, circRNA sequencing performed to analyze expression hippocampus mice.We confirmed that mmu_circ_0001442 responsive stimulation PND. The mmu_circ_0001442/miR-125a-3p/NUFIP2 axis predicted and according analysis databases, including pita, miRanda, TargetScan, miRDB, micro-CDS, PolymiRTS, TarBase v.8. Subsequently, verified qPCR double-luciferase reporter gene assays. In vitro, we found rarely had an effect on growth BV2 cells CCK-8 assay, but it attenuated cytotoxic lipopolysaccharide (LPS) cells. addition, LPS promoted release proinflammatory cytokines, IL-6 TNF-α, cells, whereas knockdown NUFIP2 partially abrogated this effect.Taken together, inhibited damage neuroinflammation alleviate disorders via activation axis. This study provides a novel rationale for developing potential therapeutic agent prevention.

Resveratrol is a natural phenolic compound with known benefits against neurodegeneration. We analyzed in vitro the protective mechanisms of resveratrol proinflammatory monomeric C-reactive protein (mCRP). mCRP increases risk AD after stroke and we previously demonstrated that intracerebral induces AD-like dementia mice. Here, used BV2 microglia treated for 24 h presence or absence resveratrol. Cells conditioned media were collected analysis. Lipopolysaccharide (LPS) has also been implicated progression so LPS was as resveratrol-sensitive reference agent. at concentration 50 µg/mL activated nitric oxide pathway NLRP3 inflammasome pathway. Abstract Autism spectrum disorder (ASD) is a neurodevelopmental based on synaptic abnormalities. The estimated prevalence rate of male individuals diagnosed with ASD prevails over females in proportion 4:1. Consequently, males remain the main focus studies clinical and experimental settings. Meanwhile, some point to an underestimation this females. In work, we studied sex differences behavioral phenotypes mouse models. Juvenile female Shank3 Δ4–22 Cntnap2 −/− mutant mice their WT littermates were used experiments. animals subjected Three-Chamber Sociability Test, then euthanized, whole cortex was for evaluation phenotype. Protein levels glutamatergic (NR1) GABAergic (GAD1 VGAT) neuronal markers measured. level synaptophysin (Syp) also Dendritic spine density somatosensory neurons analyzed by Golgi staining methods. Spine Density GAD1, NR1, VGAT, Syp significantly reduced compared control group irrespective sex, indicating impaired development mice. These results consistent lack three-chamber sociability test between conclusion, both mutations undergo similar aberrations as counterparts need be along animals. Rectifying the aberrant microenvironment of a disease through maintenance redox homeostasis has emerged as promising perspective with significant therapeutic potential for Alzheimer's (AD). Herein, we design and construct novel nanozyme-boosted MOF-CRISPR platform (CMOPKP), which can maintain rescue impaired AD. By modifying targeted peptides KLVFFAED, CMOPKP traverse blood–brain barrier deliver CRISPR activation system precise Nrf2 signaling pathway downstream proteins in regions characterized by oxidative stress, thereby reinstating neuronal antioxidant capacity preserving homeostasis. Furthermore, cerium dioxide possessing catalase enzyme-like activity synergistically alleviate stress. Further vivo studies demonstrate that effectively cognitive impairment 3xTg-AD mouse models. Abstract Normal brain functioning relies on high aerobic energy production provided by mitochondria. Failure to supply a sufficient amount of energy, seen in different disorders, including autism spectrum disorder (ASD), may have significant negative impact development and support functions. Mitochondrial dysfunction, manifested the abnormal activities electron transport chain impaired metabolism, greatly contributes ASD. The aberrant this organelle is such importance that ASD has been proposed as mitochondrial disease. It should be noted not only function In particular, these organelles are involved regulation Ca 2+ homeostasis, mechanisms programmed cell death, autophagy, reactive oxygen nitrogen species (ROS RNS) production. Several syndromes originated from mitochondria-related mutations display phenotype.
