Chronic alcohol consumption can lead to live disease (ALD). Steatosis is a critical hallmark of ALD, making it an important stage for therapeutic intervention. Saikosaponin A (SSa), compound found in Radix Bupleuri, has previously shown promising hepatoprotective, anti-inflammatory, and antioxidant properties. However, its role ALD remains understudied. We employ cell-based screening models chronic-plus-binge ethanol-fed mouse model investigate the protective mechanisms SSa metabolite Saikogenin (SGA), against ethanol-induced hepatocyte injury. Our RNA-seq analysis mice unveils that primarily acts through mTOR PPAR-α signaling pathways liver. Biophysical assays loss function experiments confirm SGA directly binds modulates activity SIRT1 protein, mitigating cell injury via SIRT1-mTOR-PPAR-α axis. Furthermore, displays survival prolonging advantage compared resveratrol treating ALD. This suggests holds promise as potential agent ameliorates cellular damage likely by binding activating suppressing activity.

In this study, the release of proteins and other biomolecules into an aqueous media from two red macroalgae (Sphaerococcus coronopifolius Gelidium spinosum) was studied using eight different cell disruption techniques. Isoquercitrin, a flavonoid glycoside found in various plants, has demonstrated antioxidant, anti-inflammatory, and anticancer properties. However, its hepatoprotective effects underlying mechanisms against oxidative liver injury remain unclear. In this study, we evaluated the antioxidant of isoquercitrin using integrated silico, vitro, vivo approaches. HepG2 cells exposed to arachidonic acid (AA) iron exhibited stress-induced apoptosis, which was significantly attenuated by treatment, as evidenced increased cell viability reduced apoptosis-related protein alterations. Isoquercitrin decreased reactive oxygen species (ROS) generation preserved mitochondrial function dose-dependent manner. Molecular docking Western blot analyses revealed that activates LKB1/AMPK pathway, increasing phosphorylation AMPK downstream target ACC, thereby modulating energy metabolism reducing stress. This activation LKB1 dependent, confirmed LKB1-deficient HeLa cells. Additionally, modulated YAP signaling pathway hepatic vivo, protected mice carbon tetrachloride-induced injury, serum ALT AST levels improving histopathological features. These findings suggest exerts activating metabolic enzymes, highlighting potential therapeutic agent damage.
