The intestinal absorption of active herbal constituents plays an important role in the biomedical efficacy Traditional Chinese Medicine (TCM) formulations after oral administration. TCM compounds with low bioavailability can reach distal intestine and then interact flora, influencing botanical pharmacological effects. In this study, vitro digestion ex vivo Ussing chamber model were utilized to evaluate behavior Forsythiae Fructus-Lonicerae Japonicae Flos-containing Yinqiao Jiedu Granule (YQJDG). It was found that jejunum exhibited effects for some components formula, while other ingredients low. Through further research using a combined UPLC-MS/MS 16S rDNA sequencing technique, we studied existence reciprocal interactions between YQJDG gut microbiome. fecal fermentation results showed significantly impacted microbial community composition. markedly increased abundance beneficial microorganisms, such as Parabacteroides distasonis Streptococcus gallolyticus subsp. Macedonicus, suppressed conditional pathogens including Prevotella steorerea, Haemophilus parainfluenzae, Bacteroides. Lonicera japonica Thunb. ( L. ) is an edible-medicinal herb. While the flower buds of are commonly utilized for medicinal purposes, flowers often overlooked. However, it has been discovered that contain higher levels certain active compounds compared to buds. Despite this finding, there have no reports on potential differences in pharmacological efficacy between these compounds. Utilizing results from GC-MS and LC-MS, a total 335 differential were identified, which 247 complied with Lipinski’s Rule Five concerning properties. Among these, 101 upregulated buds, while 146 flowers. Network pharmacology analysis revealed targeted 143 185 core targets, respectively, 116 being duplicates. The target proteins among duplicate targets primarily involved pathways related cancer, lipid atherosclerosis, hepatitis B, proteoglycans Alzheimer’s disease. Meanwhile, hub enriched distinct associated human T-cell leukemia virus 1 infection, focal adhesion, thyroid hormone signaling pathway, fluid shear stress atherosclerosis. Molecular docking indicated exhibited strong binding affinity targets. The total iridoid glycosides (TIG) from Lonicera japonica Thunb. were firstly enriched by D316 anion exchange resin, using the response surface method (RSM) model to optimise enrichment parameters, leading optimised RSM parameters of 1.25 g sample weight, 8.83 h adsorption time, 3.65 BV elution volume.
