Despite advances in anaerobic digestion (AD), full-scale implementation faces significant challenges, particularly during the start-up phase, where inoculum selection is crucial. This study examines impact of choice on operational and economic performance thermophilic digesters phase. Methanogenic reactors R3 R4 were inoculated with digested sludge (DiS) diluted sewage (DSS), respectively, fed hydrolyzed source-sorted organic fraction municipal solid waste (SS-OFMSW) thickened sludge, which processed R1 R2, serving as acidogenic reactors. A two-stage AD configuration was employed to mitigate inhibitory effects associated undigested (DSS). approach enabled establishment methanogenic activity when system initiated DSS. However, outperformed R4, achieving 49 % feedstock's theoretical methane potential compared 15 R4. Methane production volatile solids (VS) processing costs 18 3 times higher than R3, respectively. R3's superior attributed DiS's diverse bacterial community, over 66 genera involved hydrolysis, fatty acid production, syntrophic production. In contrast, DSS dominated by Trichococcus Lactococcus (75.4 %), primarily butyrate oxidation lactate provides valuable insights into effective for digesters.
