The anthranilic diamide insecticide chlorantraniliprole has been extensively applied to control Lepidoptera pests. However, its overuse leads the development of resistance and accumulation residue in environment. Four P450s (CYP6CV5, CYP9A68, CYP321F3, CYP324A12) were first found be constitutively overexpressed an SSB CAP-resistant strain. It is imperative further elucidate molecular mechanisms underlying P450s-mediated CAP for mitigating environmental contamination. Here, we heterologously expressed these four insect cells evaluated their abilities metabolize CAP. Western blotting reduced CO difference spectrum tests showed that P450 proteins had successfully Sf9 cells, which are indicative active functional enzymes. recombinant CYP6CV5, CYP324A12 exhibited a preference metabolizing fluorescent model probe substrates EC, BFC, EFC, EC with enzyme activities 0.54, 0.67, 0.57, 0.46 pmol/min/pmol P450, respectively. In vitro metabolism revealed distinct metabolic rates (0.97, 0.86, 0.75, 0.55 P450) efficiencies (0.45, 0.37, 0.30, 0.17) enzymes, thereby elucidating different protein catalytic activities. Furthermore, docking confirmed differences unveiled hydroxylation reaction generating N-demethylation methylphenyl during metabolism. Our findings not only provide new insights into at level but also demonstrate significant capacities multiple degradation facilitate evaluation mitigation toxic risks associated application

Abstract Background Cyproflanilide is a novel chemical that already undergoing insecticide registration in China and has been categorized as member of group 30 by the IRAC. Since it was first detected 2019, fall armyworm (FAW), Spodoptera frugiperda , become serious pest China. Our laboratory field efficacy trials indicated cyproflanilide exhibits high larvicidal activity against FAW. However, effect FAW remains unknown. And worth exploring further before becomes commercially available. Results We found larvae exposed to had significantly shorter body length higher death rates compared control larvae. Additionally, we surviving longer developmental period The potential molecular mechanisms were investigated using comparative transcriptomic analyses on larval samples subjected three treatments, including two other commonly used insecticides China, chlorantraniliprole avermectin. several subunits γ-aminobutyric acid receptor (GABAR), possible target protein cyproflanilide, up-regulated at transcriptional level during cyproflanilide-induced stress. between cyproflanilide-treated samples, identified 131 differentially expressed genes (DEGs) associated with detoxification metabolism. Of these, four P450 under stress but not DEGs when avermectin, or 23 pesticides from previous reports. Furthermore, discovered an interesting gene aggregation region for insect cuticle proteins (CPs) 18 th chromosome, which likely related cross-resistance Conclusions results contribute greater understanding affects similarities differences profiling insecticides.

In response to pathogen infection, some plants increase production of secondary metabolites, which not only enhance plant defense but also induce fungicide resistance, especially multidrug resistance (MDR) in the through preadaptation. To investigate cause MDR Botrytis cinerea, grapes 'Victoria' (susceptible B. cinerea) and 'Shine Muscat' (resistant were inoculated into seedling leaves with followed by extraction metabolites from on days 3, 6, 9 after inoculation. The extract was analyzed using gas chromatography/quadrupole time-of-flight mass (GC/QTOF) combined solid-phase microextraction (SPME) for volatile nonvolatile metabolomic components. Nonvolatile γ-aminobutyric acid (GABA), resveratrol, piceid, carbohydrates or amino acids, coupled β-ocimene, α-farnesene, caryophyllene, germacrene D, β-copaene, alkanes, accumulated at a higher level grape infected cinerea compared noninoculated leaves. Among established metabolic pathways, seven had greater impacts, including aminoacyl-tRNA biosynthesis, galactose metabolism, valine, leucine, isoleucine biosynthesis. Furthermore, isoquinoline alkaloid biosynthesis; phenylpropanoid monobactam tropane, piperidine, pyridine phenylalanine metabolism; glucosinolate biosynthesis related antifungal activities. Based liquid (LC/QTOF) detection bioassay, infection induced (PSMs) eugenol, flavanone, reserpine, salicylic acid, all have inhibitory activity against cinerea. These compounds promoted overexpression ATP-binding cassette (ABC) transporter genes, are involved induction

Abstract Methoxyfenozide is an insecticide with a unique mode of action on the insect ecdysone receptor and has been registered for control pests all over world. In present work, Spodoptera frugiperda was exposed to sublethal lethal concentrations methoxyfenozide determine its impact specific biological traits, metabolic enzyme activity, expression detoxification enzymes. The result showed that 72-h posttreatment LC50 LC70 significantly reduced fecundity (eggs/female) F0 generation compared those group. However, duration prepupal period increased. exposure LC10, LC30, LC50, extended developmental larvae in F1 individuals. decreased, population life table parameters were also affected. activity carboxylesterases little significant change, whereas glutathione S-transferases (GSTs) cytochrome P450 monooxygenases (P450s) altered after methoxyfenozide. total, 24-, 48-, 96-h could cause upregulation genes such as CYP6AE44, CYP6B39, CYP9A26, CYP9A58, CYP9A59, CYP9A60, well GST including GSTe3, GSTe9, GSTe10, GSTe15, GSTo2, GSTs1, GSTs5, GSTm2, GSTm3. These findings be instrumental elucidating molecular mechanisms underlying effects S. frugiperda.

Silkworm is a typical monophagous insect that can only feed on fresh mulberry leaves. The mechanism for this nature not fully understood. One bitter gustatory receptor (GR) GR66 located the maxilla of mouthpart has been reported to be an important factor influencing feeding preference silkworm. However, mutants nonhost plant was very low, suggesting other factors related silkworm monophagy need further explored. In study, 10 GRs were screened out based their specific high expression in silkworm, and 4 most expressed (GR15, GR43, GR69, GR66) knocked by clustered regularly interspaced short palindromic repeats (CRISPR) / CRISPR-associated nuclease 9. Feeding experiments showed except GR69-/-, host range GR43-/-, GR15-/-, GR66-/- expanded remarkably, both GR43-/- revealed highest plants. Moreover, strict leaves almost eliminated double mutant silkworms GR66-/-. These results imply it likely variety different are involved determining larvae. addition, GR15 mutation significantly inhibited ovarian development, resulting significant decrease oviposition. findings enhance our understanding provide possibility molecular breeding polyphagous future.

The cotton leafworm, Spodoptera littoralis (Lepidoptera: Noctuidae), is a major agricultural pest affecting crops like cotton, maize, tomatoes, and wheat across southern Europe, Africa, the Middle East, western Asia. Whole genome analyses have revealed adaptive evolution in chemosensation detoxification genes S. littoralis. However, extent of intraspecific diversity influenced by recent evolutionary forces remains unclear. In this study, we conducted population genomics analysis using 31 individuals from sub-Saharan northern Europe to assess existence divergence identify underlying forces. We show whole differentiation between populations based on geographic origin analyzed samples. Phylogenetic indicate that European share common ancestor, distinct several African populations. FST dXY statistics along chromosomes reveal loci with restricted gene flow among These are associated population-specific selective sweeps, indicating role divergent natural selection limiting flow. Notably, these enriched genes, including cytochrome P450, multidrug resistance, xanthine dehydrogenase all which potentially detoxification. results demonstrate limits geographically possibility involvement as key trait. argue genetic heterogeneity can be considered monitoring management, strategies tailored specific may not relevant for others.
