Arctium lappa L. is a medicinal edible homologous plant, commonly known as burdock or bardana, which belongs to the Asteraceae family and abundant all over world. Genetic diversity assessment essential for A. germplasm resource conservation breeding. The techniques include morphological, biochemical, DNA marker analysis. However, limited number of available markers insufficient conduct related genetic studies. In this study, we conducted RNA sequencing cultivar 'Yanagikawa Ideal' developed SSR characterize population structure 56 accessions 8 wild relative accessions. A total 4,851 simple sequence repeats (SSRs) loci were identified. proportions mono-, di- tri-nucleotide repeat motifs 30.40%, 21.50% 33.10%, respectively. We verified reliability 28 core primer pairs through electronic polymerase chain reaction (ePCR) PCR amplification process. polymorphism information content (PIC) values ranged from 0.246 0.848, with 14 primers displaying high (PIC > 0.5). showed 100% mobility in tomentosum Miller 96.43% Synurus deltoides (Aiton) Nakai, indicating their versatility. average Shannon index (I) was 1.231, observed heterozygosity (Ho) 0.132, expected (He) 0.564. 64 divided into three clusters at distance 0.558. AMOVA analysis shows 83% variation within populations 17% among populations, highlighting implications breeding strategies.
