Sitophilus zeamais is a highly destructive pest of stored grains worldwide. To develop novel RNA insecticides, the double-stranded S. cytochrome c oxidase (dsSzCOXs: dsSzCOXI, dsSzCOXII, and dsSzCOXIII) was successfully expressed in pET28a-BL21(DE3)-RNaseIII- system remarkably downregulated expression level SzCOXs through injection or feeding methods, leading to high mortalities both larvae adults. Under their optimal conditions (isopropyl β-d-thiogalactoside, 0.6 mM), yields dsSzCOXI (at 28 °C for 4 h), dsSzCOXII 37 6 dsSzCOXIII h) were 8.49, 5.80, 6.04 μg/mL respectively. The chitosan-based gene delivery composed dsSzCOXs (CS@dsSzCOXs) established insecticidal effect adults via mixing wheat seeds, with mortality reaching 50% on day ten. Consequently, this nanoparticle-mediated expected be an effective approach control.

The fall armyworm, Spodoptera frugiperda, a lepidopteran pest from the family Noctuidae, has become major invasive since 2016. Using RNAi methods to control S. frugiperda is currently under investigation. This study first target V-ATPaseD gene of using RNAi. Injection dsRNA-V-ATPaseD into hemolymph 4th-instar larvae significantly suppressed expression at 24 and 48 h post-injection. Treated showed delayed development reduced pupation after 7 days. Subsequently, silencing was achieved through topical or oral administration chitosan/dsRNA-V-ATPaseD nanoparticles. Larvae fed these nanoparticles exhibited significant reductions in mRNA 72 h, persisting until 96 before normalizing. Additionally, treated displayed disrupted molting impaired pupation. Furthermore, were more susceptible emamectin benzoate–lufenuron LC30 concentrations, resulting 68% mortality—27% higher than pesticide alone—72 post-exposure. Combining with enhanced efficacy, providing new insights reduction sustainable for this species.

Sugar transporters play important roles in controlling carbohydrate transport and are responsible for mediating the movement of sugars into cells numerous organisms. Chromosome 1 was found possess highest number genes, harboring 15. gut, salivary glands, fat body, ovary different tissues enriched with gene expression. expression levels NlST2, 4, 7, 20, 27, 28, 31 higher gut than other tissues. When expressed Saccharomyces cerevisiae hexose transporter deletion mutant (strain EBY.VW4000), ApST4 (previously characterized) NlST4, glucose fructose, resulting rescue yeast mutant.
