Sarcosphaera coronaria, a poisonous mushroom safe for consumption when cooked properly, was investigated cytotoxic metabolites against HeLa (cervical), HT-29 (adenocarcinoma), and MCF-7 (breast) cancer cell lines. Polarity-based extracts from raw baked parts were screened, advancing acetone methanol detailed mycochemical analysis. Four new compounds isolated the extract, including three amides: N-dodecanoyl-N-(ethylsulfinyl) hexadecanamide (1), N-(1,3,4-trihydroxyoctadecan-2-yl) tetracosanamide (2), N-(1,3,4-trihydroxycosane-2-yl) (3) 1,2-dilinolenoyl glycerol (4). Compounds 1 2 obtained mushroom, while 3 4 mushroom. Structural elucidation achieved via spectroscopic techniques, IR, 1D 2D NMR, MS. Compound exhibited significant activity lines with IC50 values of 52.44 ± 2.1 71.51 0.8 µg/mL, respectively, showed moderate HT29 line (IC50: 86.85 1.7 µg/mL). similar 83.83 1.5 84.27 3.2 µg/mL), MCF7 85.91 86.41 2.7 90.91 3.3 93.74 1.48 µg/mL µg/mL) lines, respectively. Due to presence these bioactive components, has potential be developed into valuable source nutraceuticals.

Hyperuricemia (HUA), characterized by elevated serum uric acid (UA) levels, is a key risk factor for gout. In human purine metabolism, approximately 70% of UA excreted via the kidneys, while remaining 30% eliminated through intestines. Thus, intestinal microbiota plays crucial role in regulating metabolism gut–kidney axis. However, detailed mechanisms which reduces levels and supports kidney health remain unclear. this study, researchers investigated potential Lacticaseibacillus paracasei LT12, strain exhibiting xanthine oxidase (XO) inhibition activity ability to degrade inosine guanosine, reducing hyperuricemia mouse model. was induced gavaging mice with 300 mg/kg potassium oxonate hypoxanthine two weeks. The subsequent 4-week intervention included five groups: normal control group, model positive group receiving allopurinol (5 body weight), low-dose LT12 (1.5 × 10⁶ CFU/kg), high-dose (4.5 10⁹ CFU/kg). results demonstrated that L. effectively reduced inhibited hepatic XO activity, regulated renal transporter proteins (OAT1, URAT1, GLUT9, ABCG2), abundance pathogenic bacterium Corynebacterium stationis both groups. Notably, only significantly increased gut butyrate levels. conclusion, shows promise as probiotic ameliorating hyperuricemia. Future clinical studies are needed validate its efficacy.

MPEA fraction showed a better NO effects than that MPE MPH, while MPH stronger those MPE, MPM TNF-α experiment. From most active (MPEA), eight (1-8) isolated column chromatography. Of which, quercetin (3), quercitrin (7), kaempferol (2), Luteolin 7 glucoside (8) potential anti-inflammation suppressing oxidase. Conclusions exhibited significant anti-inflammatory, antioxidant, showing it promising source developing medications targeting inflammation elevated levels. Its potentially pharmaceuticals conditions like gout other inflammatory illnesses.

Bioactive peptides from food sources offer a safe and biocompatible approach to enzyme inhibition, with potential applications in managing metabolic disorders such as hyperuricemia gout, conditions linked excessive xanthine oxidase activity. Using machine learning-based screening inspired by the bioactivity of natto, two peptides, ECFK FECK, were identified Bacillus subtilis proteome validated inhibitors IC50 values 37.36 71.57 mM, respectively. Further experiments confirmed their safety through cytotoxicity assays, electronic tongue analysis demonstrated mild sensory properties, supporting edibility. Molecular dynamics simulations revealed that these stabilize critical regions, showing higher dissociation energy barrier (52.08 kcal/mol) than FECK (46.39 kcal/mol), indicating strong, stable interactions.
