Highly hygroscopic pharmaceutical and nutraceutical solids are prone to significant changes in their physicochemical properties due chemical degradation and/or solid-state transition, resulting adverse effects on therapeutic performances shelf life. Moisture absorption also leads excessive wetting of the solids, causing difficult handling during manufacturing. In this review, four formulation strategies that have been employed tackle hygroscopicity issues oral solid dosage forms pharmaceuticals/nutraceuticals were discussed. The (1) film coating, (2) encapsulation by spray drying or coacervation, (3) co-processing with excipients, (4) crystal engineering co-crystallization. Film coating work acting as barriers between active ingredients core environment, whereas excipients works mainly adding deflect moisture away from ingredients. Co-crystallization altering packing arrangements introducing stabilizing co-formers. For pharmaceuticals, co-crystallization most commonly strategies, popular for nutraceuticals (e.g., medicinal herbs, protein hydrolysates). Encapsulation is rarely applied just used nutraceuticals. Therefore, there potential improved reduction exploring beyond traditionally strategy.

Recently, Natural Deep Eutectic Solvents (NaDES) have emerged as potential solvents for boosting drug bioavailability. In this work, the mechanism of solubility enhancement some APIs belonging to BCS class II (tolbutamide, nimesulide, domperidone and cinnarizine) in these eutectic bio-solvents was investigated order get deeper insights into molecular interactions between NaDES components selected drugs. Different formulations based on choline chloride, proline, solid organic acids (citric, tartaric malic acid), sugars (glucose xylitol) water were prepared by mild heating (70 °C). Characterization unloaded (pH, Karl Fisher titration, viscosity FTIR analysis) indicated that type Hydrogen Bond Acceptor (HBA) Donor (HBD), their molar ratio well amount strongly affect extent H-bonding interactions. Hard gelatin capsules filled with maintained integrity until 6 months, proving all molecules participate H-bond network. APIs' significant respect buffer solutions (pH 1.2 6.8). Analysing having Choline HBA, it found smaller increased using larger HBD, while higher weight can be better inserted network formed HBD. NOE experiments demonstrated formation a robust supramolecular structure among protons choline, those acid water. addition, 1D ROESY spectra revealed first time crucial role (methyl groups) establishing hydrophobic relative aliphatic or aromatic portion These data suggest complex API-NaDES assembly underline is dependent balance H-bonds well. Understanding API essential use effective solubilisation aid.

Summary A review paper on the natural deep eutectic solvents (NaDES) application to extraction of bioactives from products and agri‐food wastes is introduced. NaDES are mixtures two or more compounds forming a liquid phase at room temperature based in hydrogen bond acceptors (quaternary amines, alcohols) donors (sugars, organic acids). NADES have unique properties such as low toxicity, biodegradability high solubility for wide range compounds. They represent sustainable alternative traditional variety applications, particularly metabolites extraction. Therefore, status displayed terms their inspiration nature, preparation physicochemical properties, well conditions biologically important phytochemicals alkaloids, carotenoids, phenolics (anthocyanins, isoflavonoids, curcuminoids). Remarks emerging technologies therapeutic, hydrophilic, hydrophobic systems inserted. As can be easily prepared renewable sources, they promising green food, nutraceutical, pharmaceutical industries.
