Purpose: Atopic dermatitis (AD) is the most common chronic inflammatory skin disease that severely impairs patient's life quality and represents significant therapeutic challenge due to its pathophysiology arising from barrier dysfunction.Topical corticosteroids, mainstay treatment for mild moderate AD, are usually formulated into conventional dosage forms impeded by low drug permeation, resulting in high doses with consequent adverse effects, also lack properties would strengthen barrier.Herein, we aimed develop biomimetic lamellar lyotropic liquid crystals (LLCs), offering a novel alternative AD treatment.Methods: In screening studies, pseudoternary phase diagrams alongside polarized light microscopy (PLM) viscosity measurements were utilized.Next, selected LCCs underwent comprehensive characterization via PLM, small-angle X-ray scattering, differential scanning calorimetry, rheological analysis.Lastly, their performance was evaluated compared commercially available reference medicine chemical stability study, vitro permeation testing, safety assessment using cell proliferation assay, inverted microscopy, Raman mapping of keratinocytes, besides gap closure assay performed live-cell imaging.Results: Formulation (L/T)Ho30, containing highest amount lecithin/Tween 80 mixture (21%) hempseed oil (28%), demonstrated microstructure hydration potential favourable features administration.Moreover, comparison medicine, it stood out providing suitable BD (betamethasone dipropionate) stability, improved 3-fold excellent biocompatibility over 85% at all tested concentrations, ensuring keratinocytes' integrity, as well promoting healing observed after 36 hours.Conclusion: Unique multi-target delivery strategy depicted newly developed bioinspired structurally resembling stratum corneum intercellular lipids, incorporated drug, composed multifunctional components synergistically barrier, presented here shows promising approach treatment.

Responsive nanostructured films from food-grade lipids can be valuable for food, pharmaceutical, and biotechnological science. Lyotropic liquid crystalline structures that respond to enzymes in their environment can, instance, innovated as drug delivery platforms or biosensors.
