β-hydroxybutyrate (β-HB) may serve as a signaling metabolite in many physiological processes beyond fuel source for tissues. The present study aims to investigate the role of lysine β-hydroxybutyrylation (Kbhb) modification mediated by β-HB regulating metabolic homeostasis both vivo vitro. starvation ketosis type 1 diabetes mouse models were introduced evaluate influence on Kbhb mice. modifications OXCT1 HMGCS2, two rate-limiting enzymes ketogenesis utilization, showed positive correlation with level vitro vivo. levels increased during fasting but decreased after refeeding. all detected tissues minor change since blood non-significantly model. experiments further indicated that mutation at site significantly inhibited enzymatic activity not HMGCS2. SIRT1 CBP identified potential dehydrogenase transferase OXCT1, respectively. Lysine 421 increases its enzyme accumulation, accelerating utilization finally maintaining homeostasis.
