The mycotoxin ochratoxin A (OTA) causes nephrotoxicity, hepatotoxicity, and immunotoxicity in animals humans. farnesoid X receptor (FXR) is a member of the NR family highly expressed kidney, which has an antilipid production function. Ferroptosis iron-dependent form regulated cell death involved several pathophysiological kidney injury. present study aims to evaluate role FXR ferroptosis OTA-induced nephrotoxicity mice HK-2 cells. Results showed that OTA induced as demonstrated by inducing histopathological lesions neutrophil infiltration increasing serum BUN, CRE, UA levels, Ntn-1, Kim-1, pro-inflammatory cytokine expression, decreasing IL-10 expression viability treatment also deficiency, ROS release, MDA level increase, GSH content decrease, 4-HNE labile iron pool lipid peroxidation levels well Acsl4, TFR1, HO-1 mRNA protein GPX4 FTH expressions, mitochondrial activator (GW4064) rescued accumulation peroxides, intracellular ROS, Fe2+, inhibited ferroptosis, alleviated nephrotoxicity. inhibitor (Fer-1) prevented attenuated Collectively, this elucidates played critical via regulation provides novel strategy against

Safflower is a traditional Chinese medicine used for treating gynaecological diseases. However, its material basis and mechanism of action in the treatment endometritis induced by incomplete abortion are still unclear. This study aimed to reveal safflower through comprehensive methods, including network pharmacology 16S rDNA sequencing.Network molecular docking methods were screen main active components potential mechanisms rats. A rat model endometrial inflammation was established. The rats treated with total flavonoids (STF) based on forecasting results, serum levels inflammatory cytokines analysed, immunohistochemistry, Western blots, sequencing performed investigate effects ingredient mechanism. The prediction results showed 20 260 targets safflower, 1007 related caused abortion, 114 drug-disease intersecting targets, TNF, IL6, TP53, AKT1, JUN, VEGFA, CASP3 other core PI3K/AKT, MAPK signalling pathways may be closely leading endometritis. animal experiment that STF could significantly repair uterine damage reduce amount bleeding. Abstract Endometritis is mostly caused by childbirth or postpartum uterine infection. It one of the important reasons leading to female infertility. Caffeic acid (CA) and its derivatives are widely found in some foods traditional Chinese medicine, have biological activities such as antioxidant, free radical scavenging, anti‐inflammatory, anti‐infection. In this study, we aimed explore effect CA on Staphylococcus aureus ‐induced endometritis. The contents TNF‐α IL‐1β were detected ELISA S. endometritis model. Western blot assay was used detect expression AMPKα/mTOR/HIF‐1α pathway related proteins GPX4 expression. addition, concentrations MDA, GSH, iron tested kits. Compared with model group, treatment significantly alleviated injury, MPO activity, inflammatory factors IL‐1β, NF‐κB activation. Meanwhile, inhibited ferroptosis, confirmed decreased MDA concentration up‐regulated GSH level. Furthermore, attenuated HIF‐1α phosphorylated mTOR increased AMPK conclusion, inhibits inflammation ferroptosis regulating signalling alleviate mice.
