Chronic pain and mental health disorders, such as depression anxiety, frequently co-occur share underlying mechanisms involving neuronal excitability synaptic transmission. The inwardly rectifying potassium channel 4.1 (Kir4.1), predominantly expressed in glial cells, is crucial for maintaining extracellular glutamate homeostasis. Dysregulation of Kir4.1 leads to altered activity, contributing both chronic disorders. In pain, downregulation impairs buffering clearance, increasing enhancing signaling through peripheral central sensitization. impaired function disrupts neurotrophic factor secretion neuroinflammatory pathways, leading mood disturbances. This review primarily summarizes findings from preclinical studies examine the relationship between pathogenesis discussing its molecular structure, expression patterns, functional roles. Endometriosis, a prevalent chronic gynecological condition, is frequently associated with infertility and pelvic pain. Despite numerous studies indicating correlation between epigenetic regulation endometriosis, its precise genetic etiology remains elusive. Methyltransferase-like 14 (METTL14), crucial component of the N6-methyladenosine (m6A) RNA methyltransferase complex an binding scaffold, known to play pivotal role in various human diseases. The possibility that single nucleotide polymorphisms (SNPs) METTL14 gene contribute susceptibility endometriosis has not been thoroughly investigated. Postoperative cognitive dysfunction (POCD) is a common complication after total knee arthroplasty (TKA), impacting recovery and quality of life. This study aims to investigate central sensitization (CS) as an independent risk factor for POCD improve preoperative screening postoperative interventions. A retrospective analysis was conducted on 142 TKA patients from January 2020 May 2024 across three hospitals. Data were collected at six time points: preoperatively (T0), intraoperatively (T1), postoperatively days 1 (T2), 3 (T3), 7 (T4), 30 (T5). Patients classified into CS (CSI ≥ 40) non-CS < groups according Central Sensitization Inventory (CSI) score. Cognitive function incidence assessed with the Mini-Mental State Examination (MMSE), Knee Injury Osteoarthritis Outcome Score (KOOS). Logistic regression used identified factors POCD. ABSTRACT Fatty acid binding protein 7 (FABP7) is prominently expressed in astrocytes and a critical regulator of inflammatory responses. Accumulating evidence suggests that FABP7 crucial neuropsychological disease through the modulation spinogenesis. Nonetheless, impact on depressive disorders underlying mechanisms not fully understood. Here, we investigated antidepressant properties using chronic unpredictable mild stress (CUMS)‐induced model depression possible mechanisms. Our results revealed depressive‐like behavior induced by CUMS was associated with decreased levels hippocampus (HP). Furthermore, overexpression HP mitigated increased expression its downstream target caveolin‐1 (Cav‐1). specifically regulates astrocyte marker GFAP, as well blood–brain barrier (BBB)‐associated proteins AQP4, CLDN‐5, occludin, LRP1. Notably, CUMS‐induced upregulation pro‐inflammatory factors IL‐1β IL‐6 also significantly reversed HP. This intervention led to postsynaptic proteins, including PSD95 GluA1, an increase brain‐derived neurotrophic factor (BDNF) enhanced neuronal dendritic spine density.
