Abstract Sex differences in pain sensitivity have contributed to the fact that medications for curing chronic are unsatisfactory. However, underlying mechanism remains be elucidated. Brain‐derived estrogen participates modulation of sex and related emotion. G protein‐coupled receptor 30 (GPR30), identified as a novel with different distribution than traditional receptors, has been proved play vital role regulating affected by estrogen. contribution its sexually dimorphic pain‐related behaviors not fully explored. In current study, immunofluorescence assays were applied mark neurons expressing GPR30 male female mice (in metestrus proestrus phase) brain regions. The express CaMKIIα or VGAT also labeled observe overlap GPR30. We found females had more GPR30‐positive (GPR30 + ) primary somatosensory (S1) insular cortex (IC) males. lateral habenula (LHb) nucleus tractus solitarius (NTS), males females. Moreover, within LHb, expression varied estrous cycle phase; fewer those proestrus. addition, neurons, which co‐expressed medial preoptic (mPOA) males, while basolateral complex amygdala (BLA). These findings may partly explain modulatory effects emotional phenotypes between sexes provide basis comprehension sexual dimorphism GPR30, finally new targets exploiting treatments sex‐specific pain.

Comorbid anxiodepressive-like symptoms (CADS) in chronic pain are closely related to the overactivation of lateral habenula (LHb). Hyperpolarization-activated cyclic nucleotide-gated (HCN) channels have been implicated play a key role regulating neuronal excitability. However, HCN LHb during CADS has not yet characterized. This study aimed investigate effect on pain.

We take this basic approach integrate it into a framework could provide real-time measures peri-surgical period. Abstract Electroacupuncture (EA) has been successfully used to treat painful conditions. However, its analgesic mechanism remains poorly understood. Here, we established a Complete Freund's Adjuvant (CFA)-induced inflammatory pain model. After CFA injection, P2RY14 was upregulated in primary somatosensory cortex of hindlimb (S1HL). EA, not sham alleviated the nociceptive thermal hyperalgesia and reduced upregulation mice. Injection agonist uridine diphosphate glucose (UDPG) into bilateral S1HL reversed EA-induced analgesia. Our results demonstrate that analgesia CFA-induced can be achieved by downregulation S1HL.
