Ferroptosis, a new type of programmed cell death proposed in recent years, is characterized mainly by reactive oxygen species and iron-mediated lipid peroxidation differs from death, such as apoptosis, necrosis, autophagy. Ferroptosis associated with variety physiological pathophysiological processes. Recent studies have shown that ferroptosis can aggravate or reduce the occurrence development diseases targeting metabolic pathways signaling tumors, ischemic organ damage, other degenerative related to peroxidation. Increasing evidence suggests closely linked onset progression various ophthalmic conditions, including corneal injury, glaucoma, age-related macular degeneration, diabetic retinopathy, retinal detachment, retinoblastoma. Our review current research on reveals significant advancements our understanding pathogenesis, aetiology, treatment these conditions.

Prior studies have elucidated that alterations in gut microbiota are associated with a spectrum of tumors and metabolic disorders. However, the diagnostic value epithelial ovarian cancer remains insufficiently investigated. A total 34 patients diagnosis (EOC), 15 benign (TB), 30 healthy volunteers (NOR) were enrolled this study. Fecal samples collected, followed by sequencing V3–V4 region 16S rRNA gene. Chimeric antigen receptor T cell (CAR-T) therapy has shown success in treating hematological malignancies, but its effectiveness against solid tumors is hindered by exhaustion. During vitro expansion, tonic signaling induced CAR expression contributes to CAR-T exhaustion, which can be mitigated inhibiting calcium signaling. Given that sodium citrate chelate ions and inhibit signaling, this study, we investigated whether could reduce exhaustion enhance function. We constructed anti-CD70 cells cultured them the presence of citrate. The characteristics functionality citrate-pretreated were assessed through vivo experiments. To further validate our observation, also treated anti-mesothelin (MSLN) with detected phenotypes anti-tumor function cells. found exhibited reduced increased memory proportions, enhanced efficacy both vivo. Notably, treatment improved persistence prevented tumor recurrence. These beneficial effects observed anti-MSLN Transcriptomic metabolite analyses revealed inhibited mTORC1 activity, glycolysis pathways, thus modulating differentiation. Our findings suggest supplementation during expansion a promising strategy improve for preventing promoting formation.

Abstract Effective and precise treatment of breast cancer, particularly with bone metastasis, remains a significant challenge. Here, dual‐gating strategy combining locally delivered borate glass (BG) ultrasound (US) is developed for the effective inhibition cancer by targeting transient receptor potential vanilloid 2 (TRPV2). The results demonstrate that after local delivery BG to solid tumor, US effectively triggers calcium overload activating overexpressed TRPV2 channels, leading mitochondrial autophagy apoptosis in cells, thereby inhibiting tumor growth high precision. These effects are validated subcutaneous, orthotopic, TRPV2‐overexpressing mouse models. In metastasis model, combined simultaneously suppresses promotes regeneration. Overall, this offers safe efficient approach cancers expression provides new insights into design clinical translation calcium‐overload‐based therapies.
