In recent years, small interfering RNA (siRNA) has been widely used in the treatment of human diseases, especially tumors, and shown great appeal. However, clinical application siRNA faces several challenges. Insufficient efficacy, poor bioavailability, stability, lack responsiveness to a single therapy are main problems affecting tumor therapy. Here, we designed cell-penetrating peptide (CPP)-modified metal organic framework nanoplatform (named PEG-CPP33@ORI@survivin siRNA@ZIF-90, PEG-CPP33@NPs) for targeted co-delivery oridonin (ORI), natural anti-tumor active ingredient) survivin vivo. This can improve stability bioavailability efficacy monotherapy. The high drug-loading capacity pH-sensitive properties zeolite imidazolides endowed PEG-CPP33@NPs with lysosomal escape abilities. Polyethylene glycol (PEG)-conjugated CPP (PEG-CPP33) coating significantly improved uptake vitro results showed that ORI greatly enhanced effect PEG-CPP33@NPs, demonstrating synergistic between siRNA. summary, novel nanobiological platform loaded presented herein advantages cancer therapy, provides an attractive strategy chemotherapy gene

The development of new functional materials based on metal-organic frameworks (MOFs) for adsorption and catalytic applications is one the promising trends modern science. Zr-based MOFs, specifically UiO-66, are considered as supports metallic catalysts 5-hydroxymethylfurfural platform molecule reduction into valuable products. present work focused effect NH2 modification UiO-66 its structure properties. samples were prepared by a solvothermal method. obtained was studied X-ray diffraction, IR spectroscopy, UV-visible low-temperature nitrogen adsorption. Basic properties investigated HCl CH3COOH adsorption, electrokinetic electrophoretic light scattering. UiO-66-NH2 with different contents aminoterephthalate linkers successfully prepared. A gradual decrease in specific surface area fraction micropores diameter ~0.9 nm observed an increase content. proportional total number basic sites established content up to 75%. At same time, noticeable their strength higher observed.
