Abstract Extracellular vesicles (EVs) are heterogeneous membrane-like secreted by living cells that involved in many physiological and pathological processes act as intermediaries of intercellular communication molecular transfer. Recent studies have shown EVs from specific sources regulate tissue repair regeneration delivering proteins, lipids, nucleic acids to target signaling molecules. Nanotechnology breakthroughs facilitated the development exploration engineered for repair. Enhancements through gene editing, surface modification, content modification further improved their therapeutic efficacy. This review summarizes potential regeneration, mechanisms action, research progress regenerative medicine. highlights design logic typical examples explores prospects The aim this is provide new insights into applications, thereby expanding use

Schwann cells are essential for the maintenance and function of motor neurons, axonal networks, neuromuscular junction. In amyotrophic lateral sclerosis, where neuron is progressively lost, cell may also be impaired. Recently, important signaling potential trophic activities cell-derived exosomal vesicles have been reported. This case report describes treatment a patient with advanced sclerosis using serial intravenous infusions allogeneic vesicles, marking, to our knowledge, first instance such treatment. An 81-year-old male presented 1.5-year history rapidly progressive sclerosis. After initial diagnosis, underwent combination generic riluzole, sodium phenylbutyrate taurursodiol. Retinal diseases are the leading cause of blindness, resulting in irreversible degeneration and death retinal neurons. One such cell type, ganglion (RGC), is responsible for connecting retina to rest brain through its axons that make up optic nerve primary lost glaucoma traumatic neuropathy. To date, different therapeutic strategies have been investigated protect RGCs from preserve vision, yet currently available restricted treating neuron loss by reducing intraocular pressure. A major barrier identified these studies drug delivery RGCs, which large part due stability, short duration time at target, low efficiency, undesired off-target effects. Therefore, a system deal with problems needed ensure maximum benefit candidate material. Extracellular vesicles (EV), nanocarriers released all cells, lipid membranes encapsulating RNAs, proteins, lipids. As they naturally shuttle encapsulated compounds between cells communicative purposes, may be exploitable offer opportunities overcome hurdles delivery, including molecular weight, barriers retina, adverse Here, we summarize potential an EV system, discussing their superiorities application target RGCs.

This study evaluated the neuroprotective potential of peptain-1 conjugated to a cell-penetrating peptide (CPP-P1) in an ocular hypertension model glaucoma. Brown Norway (BN) rats were subjected intraocular pressure (IOP) elevation via intracameral injection silicone oil (SO), with concurrent intravitreal injections either CPP-P1 or vehicle. Retinal cross-sections analyzed for markers neuroprotection, including cAMP response element-binding protein (CREB), phosphorylated CREB (p-CREB), growth-associated protein-43 (GAP43), synapsin-1 (SYN1), and superoxide dismutase 2 (SOD2). Hematoxylin eosin staining was used assess retinal-layer thickness. SO-treated exhibited significant reductions thickness inner nuclear layer (INL, 41%, p = 0.016), plexiform (IPL, 52%, 0.0002), ganglion cell (GCL, 57%, 0.001). treatment mitigated these reductions, preserving INL by 32% (p 0.059), IPL 19% 0.119), GCL 31% 0.057). Retinal ganglion cell (RGC) degeneration leads to irreversible blindness. Müller glia (MG) play pivotal roles in retinal homeostasis and disease through paracrine signaling. Small extracellular vesicles (sEVs) are bioactive nanomaterials derived from all types of live cells recognized as a potential strategy for neuroprotective therapy. The aim this study is investigate the MG-derived sEVs (MG-sEVs) mouse model optic nerve injury (ONC). It found that MG-sEVs treatment effectively mitigates RGC suppresses microglial activation, thereby improves visual function ONC mice. transcriptomic analysis reveals strong correlation between C-x3-c motif chemokine ligand 1 (Cx3cl1)-mediated glial activation inflammation. Subsequently, it confirmed expression levels Cx3cl1 proinflammatory cytokines significantly decreased retinas treated with MG-sEVs. components cargo identifies miR-125b-5p miR-16-5p target gene regulate its expression. also observed colocalizes on microglia transgenic receptor (Cx3Cr1)-GFP In conclusion, mitigate by suppressing via Cx3cl1-Cx3cr1 This research provides additional opportunities degeneration.
