Non-alcoholic fatty liver disease (NAFLD) is a clinicopathological syndrome characterized by abnormalities in hepatic fat deposition, the incidence of which has been increasing year recent years. It become largest chronic globally and one important causes cirrhosis even primary cancer formation. The pathogenesis NAFLD not yet fully clarified. Modern medicine lacks targeted clinical treatment protocols for NAFLD, most drugs lack efficacy have high side effects. In contrast, Traditional Chinese Medicine (TCM) significant advantages prevention widely recognized scholars around world. years, through establishment "medicine-disease-target-pathway" network relationship, pharmacology can explore molecular basis role medicines from various perspectives, predicting pharmacological mechanism corresponding medicines. This approach compatible with holistic view based on pattern differentiation TCM used research. this paper, searching relevant databases such as PubMed, Web Science, Embase, we reviewed analyzed signaling pathways specific mechanisms action single medicine, combinations, patent These related studies demonstrated therapeutic characteristics multi-components, multi-targets, multi-pathways, provided strong support exact exerted clinic. conclusion, believe that more line mindset treating diseases, but some limitations. future, should eliminate potential risks false positives negatives, clarify interconnectivity between components, targets, conduct deeper or experimental studies.

Abstract Background Fatty liver hemorrhage syndrome (FLHS) becomes one of the most major factors resulting in laying hen death for caged egg production. This study aimed to investigate therapeutic effects Lactiplantibacillus plantarum ( Lp. ) FRT4 on FLHS model with a focus lipid metabolism, and gut microbiota. Results The hens was established by feeding high-energy low-protein (HELP) diet, treatment groups were fed HELP diet supplemented differential proportions FRT4. results indicated that increased rate, reduced accumulation regulating metabolism (lipid synthesis transport) improving microbiota composition. Moreover, regulated glycerophospholipid metabolism. Meanwhile, “gut-liver” axis analysis showed there correlation between metabolites. Conclusions improved performance alleviated diet-induced through axis. Our findings reveal could be underlying mechanism anti-FLHS effect future use as an excellent additive prevention mitigation hens.

Background Non-alcoholic fatty liver disease (NAFLD) is a common chronic disease, characterized by hepatic lipid accumulation. The Fufangduzhong formula (FFDZ) traditional Chinese medicine (TCM) formulation composed of Eucommia ulmoides Oliv. , Leonurus artemisia (Lour.) S. Y. Hu, Prunella vulgaris Linn Uncariarhynchophylla (Miq.) Miq. ex Havil., and Scutellaria baicalensis Georgi. It has demonstrated hepatoprotective effects the ability to reduce However, its mechanisms against NAFLD remain unclear. Methods UPLC-MS/MS was used identify FFDZ metabolites. C57BL/6J mice were fed high-fat diet (HFD) supplemented with or without (HFD+L, 0.45 g/kg/d; HFD+H, 0.9 g/kg/d) for 12 weeks. Biochemical indicators histopathological observations utilized assess extent metabolic homeostasis disorder steatosis. An analysis differentially expressed genes regulated signaling pathways conducted using transcriptomics. Metabolomics performed investigate significantly changed endogenous metabolites associated in serum UPLC-Q-TOF/MS. Western blot employed detect proteins involved metabolism-related pathways. Oleic acid-induced steatosis examine lipid-lowering effect FFDZ-containing vitro . Results A total eight active identified from through analysis. reduced body weight, levels inflammatory cytokines, it ameliorated steatosis, profiles, insulin sensitivity, glucose tolerance HFD-induced NAFLD. Transcriptomics revealed that modulated pathways, including PPAR pathway, Fatty acid metabolism, AMPK pathway. Meanwhile, indicated downregulated expression synthesis-related (Srebp-1c, Acly, Scd-1, Fasn, Acaca, Cd36) upregulated oxidation-related (p-Ampk, Ppar-α, Cpt-1). Furthermore, metabolomics FFDZ-mediated reversal phospholipid dysregulation (PC, PE, LPC, LPE). Additionally, remarkedly OA-induced accumulation HepG2 cells. Conclusion present results demonstrate exerts anti-NAFLD enhancing as well regulating Ampk pathway ameliorate metabolism disorder, lipotoxicity, responses.

The traditional prescription of Liangxue-Qushi-Zhiyang decoction (LQZ) has been demonstrated to be efficacious in treating atopic dermatitis (AD), a chronic inflammatory skin disorder marked by intense itching, redness, rashes, and thickening. Nevertheless, there an inadequate systematic exploration the potential targets, biological processes, pathways for AD treatment through LQZ. study objective was evaluate efficacy possible mechanism LQZ mice. In our study, we identified primary compounds LQZ, analyzed hub constructed network. Subsequently, predicted mechanisms were experimentally studied validated vivo, as determined network pharmacological analysis. A total 80 serum components ultra-performance liquid chromatography-electrospray ionization-tandem mass spectrometry (UPLC-ESI-MS/MS), among which 49 absorbed into bloodstream. Our results indicated that targets six putative key factors MAPK signaling pathway, play essential roles AD, namely, EGFR, p-MAPK1/3, p-MAPK14, IL-1β, IL-6, TNF-α. We observed spleen coefficient, scores, ear thickness all downregulated 2,4-dinitrochlorobenzene (DNCB)-induced mice after treatment. Histological analysis dorsal further revealed significantly decreased inflammation, epidermal thickness, mast cell numbers compared DNCB group. effectiveness reducing dermal damage mouse model AD. This study aimed to investigate the potential of electroacupuncture as an intervention for inducing 'Awakening and Opening Brain' in rats with stroke models induced by middle cerebral artery occlusion/reperfusion (MCAO/R). The efficacy alleviating ischemic injury was evaluated using Longa scores, triphenyl tetrazolium chloride staining, hematoxylin eosin staining. Non-targeted metabolomics analysis conducted identify differential metabolite changes before after treatment MCAO/R rats. Network pharmacology then performed correlate these metabolites stroke. PI3K/AKT/NF-κB signaling pathway identified a key target. In vivo experiments further validated mechanism which promotes M2 microglial polarization through inhibition demonstrated that reduces brain damage inhibits inflammation modulating promoting microglia from M1 M2.
