Scutellarin (Scu), a flavonoid from herbal Erigeron breviscapus (Vaniot) Hand-Mazz, exerts neuroprotective effects against cerebral ischemia. However, whether the of Scu are related to mitochondrial protection needs further investigation. In this study, we aimed clarify mechanisms HT22 cells injury caused by oxygen-glucose deprivation and reperfusion (OGD/R). Our results proved that significantly reduced overload intracellular reactive oxygen species (cellar ROS) mitochondria (mito-ROS), ameliorating oxidative stress damage. TUNEL positive rate, Caspase-3 activity, Cytochrome c (Cyto-c) expression remarkably decreased following treatment. Meanwhile, could maintain morphology reverse ultrastructure changes. And membrane potential (MMP), consumption rate (OCR), adenosine triphosphate (ATP) production Na

Angiogenesis, integral to the pathogenesis of rheumatoid arthritis (RA), contributes advancement synovial hyperplasia, pannus formation, and bone destruction. The precise effect Huayu Tongbi formula (HT), a proven traditional Chinese medicine for RA treatment, on angiogenesis remains uncertain. To investigate effects HT potential underlying molecular mechanisms. target spectrum pharmacological were screened using network pharmacology. In vitro, RA-FLS stimulated with CoCl2 simulate hypoxic conditions. Human umbilical vein endothelial cells (HUVECs) cultured conditioned medium (CM) from supplemented or HIF1A inhibitor LW-6 impact cell secretions angiogenesis, encompassing proliferation, migration, adhesion, lumen formation. Western blot was employed assess protein levels hypoxia-inducible factor 1 subunit alpha (HIF1A), vascular growth receptor 2 (VEGFR2), phosphorylated VEGFR2 (p-VEGFR2). Enzyme-linked immunosorbent assay (ELISA) utilized quantify VEGFA secretion, while quantitative real-time PCR (qRT‒PCR) measured mRNA expression VEGFA, Angiopoietin (ANGPT1), ANGPT2. vivo, collagen-induced (CIA) model established. Pathological changes vessel counts observed via hematoxylin-eosin (HE) staining, structure evaluated X-ray, mature immature vessels classified immunofluorescence, mechanism further verified immunohistochemistry ELISA. Liquid chromatography‒mass spectrometry (LC‒MS) identify active components drug-containing serum, metabolites identified by bioinformatics analysis. Network pharmacology analysis indicated targets in correlating VEGF pathways. reversed adhesion lines suppressed luminal formation HUVECs. (10 % 15 %) effectively reduced expression, attenuated its phosphorylation, downregulated ANGPT1, ANGPT2 levels. mitigated inflammation destruction, (CD31+/α-SMA-) (CD31+/α-SMA+) synovium, inhibited HIF1A-VEGFA-ANGPT axis. Finally, 19 serum successfully identified, we also unexpectedly discovered that might affect metabolic pathways such as proline tyrosine metabolism, which have been reported be involved angiogenesis. has promising alleviating at least part through axis inhibition, reduces cross-talk between Our results suggest is drug candidate treatment
