Introduction Prolonged hyperglycemia caused by type 2 diabetes mellitus (T2DM) can lead to liver injury and disrupt the community of gut microbiota that pose significant risks human health. As a food rich in variety active ingredients, Zingiber striolatum ( Z. ) exhibits hypoglycemic hypolipidemic effects. However, regulatory influence ethanol extract (ZSE) on T2DM mice or its potential relationship with pathology remains unexplored. Methods After one-week acclimation period, 12 were randomly selected as normal group. The remaining 48 employed model, then assigned four groups: model group, low-dose ZSE group (ZSE-L, 100 mg/kg/day), high-dose (ZSE-H, 300 positive control treated metformin hydrochloride (MET, mg/kg/day). Results 4-week intervention, results revealed significantly ameliorated fasting blood glucose (FBG), area under curve oral tolerance test (AUC OGTT) glycated serum protein (GSP) mice. Moreover, (ZSE-H) treatment increased relative abundance beneficial bacteria such Faecalibaculum , while reducing harmful Bilophila thereby alleviating insulin resistance. Additionally, ZSE-H demonstrated superior efficacy over (ZSE-L) improving FBG, AUC OGTT, other parameters. Predictive analysis correlation between parameters identified Dubosiella Bacillus Mailhella microbial biomarkers for further investigation into pathogenesis T2DM. Conclusion plays pivotal role mitigating through modulation intestinal communities.

Plantamajoside (PMS) has shown potential in mitigating cell damage caused by high glucose (HG) levels. Despite this, the precise therapeutic effects of PMS on type 2 diabetes mellitus (T2DM) and underlying regulatory mechanisms require further exploration. To investigate T2DM mice elucidate its action through vivo vitro experiments. An model MIN6 cells was established using HG palmitic acid (PA). PMS's protective effect assessed. Next, transcriptomics employed to examine how treatment affects gene expression cells. Furthermore, protein processing endoplasmic reticulum apoptosis pathways validated. A mouse used validate vivo. intervention ameliorated injury + PA-induced damage. Transcriptomic analysis revealed that were enriched treated with PMS, significant downregulation Dnajc1. Further validation indicated significantly inhibited apoptosis-related factors (Bax, CytC) stress (ERS)-related [ATF6, XBP1, Ddit3 (CHOP), GRP78], while promoting Bcl-2 Additionally, inhibitory ERS abolished upon Dnajc1 silencing. experiments demonstrated effectively improved pancreatic damage, suppressed CytC), ERS-related a mice. could alleviate tissue effectively. The mechanism involves activation, which subsequently inhibits ERS, ameliorating β-cells.

Cardiometabolic diseases (CMDs), encompassing cardiovascular and metabolic dysfunctions, characterized by insulin resistance, dyslipidemia, hepatic steatosis, inflammation, have been identified with boosting morbidity mortality due to the dearth of efficacious therapeutic interventions. In recent years, studies shown that variations in gut microbiota its own metabolites can influence occurrence CMDs. Intriguingly, composition function are susceptible exercise patterns, thus affecting inflammatory, immune, responses within host. this review, we introduce key mechanisms intestinal microecology involved onset development CMDs, discuss relationship between microecology, then analyze role beneficial effects on aiming at elucidating gut-heart axis mediated protective effect building avenues for application management
