Lomefloxacin (LOM) is an efficacious antibiotic that might lead to liver damage and other adverse reactions if consumed over extended period. Hence, this study proposed a DNAzyme-mediated DNA walker as signal amplifier in combination with substantial quantity of Au nanoparticle (NP)-loaded hydrogen-bonded organic frameworks (HOFs) surface-enhanced Raman scattering (SERS) substrate accomplish the highly sensitive detection LOM food. Crucially, due their high specific surface area strong adsorption capacity, HOFs act ideal for situ growth large amount NPs, which exhibit robust uniform SERS enhancement. Once target was presented, DNAzyme activated start produce single abundance, further triggered catalytic hairpin assembly (CHA) cycle. In addition, multi-interstitial core-shell structure Au@Ag@4-nitrobenzenethiol (4-NTP)@Au@Hp3 produced more "hot spots" significantly amplified local electromagnetic field, HOF@Au synergistically strengthening 4-NTP. Using principle, sensor achieved ultrasensitive through "signal on" strategy, limit low 4.62 × 10-14 mol/L.
