Introduction Mesenteric lymph nodes (MLNs) are central in immune anatomy. MLNs associated with the composition of gut microbiota, affecting system and system. Gut microbiota was found to differ among individuals different social hierarchies. Nowadays, excision is more frequently involved gastrointestinal surgery; however, potential side effects on dominance still unknown. Methods were removed from male mice (7–8 weeks old). Four after MLN removal, test performed investigate dominance; hippocampal serum interleukin (IL)‐1β, IL‐10, tumor necrosis factor‐alpha (TNF‐α) investigated; histopathology used evaluate local inflammation ileum. The then examined understand possible mechanism, finally intraperitoneal injection IL‐10 validate effect dominance. Results There a decrease operation group compared control group, as well levels, but no difference IL‐1β TNF‐α ileum removal. 16S rRNA sequencing analysis showed that relative abundance class Clostridia decreased group. This positively levels. Furthermore, subset increased Conclusions Our findings suggested contributed maintaining dominance, which might be reduced imbalance specific flora microbiota.

Abstract Background Triclosan [5-chloro-2-(2,4-dichlorophenoxy) phenol, TCS], a common antimicrobial additive in many personal care and health products, is frequently detected human blood urine. Therefore, it has been considered an emerging potentially toxic pollutant recent years. Long-term exposure to TCS suggested exert endocrine disruption effects, promote liver fibrogenesis tumorigenesis. This study was aimed at clarifying the underlying cellular molecular mechanisms of hepatotoxicity effect initiation stage. Methods C57BL/6 mice were exposed different dosages for 2 weeks organ toxicity evaluated by various measurements including complete count, histological analysis quantification. Single cell RNA sequencing (scRNA-seq) then carried out on TCS- or mock-treated mouse livers delineate TCS-induced hepatotoxicity. The acquired single-cell transcriptomic data analyzed from aspects differential gene expression, transcription factor (TF) regulatory network, pseudotime trajectory, communication, systematically dissect events after exposure. To verify fibrosis, expression levels key fibrogenic proteins examined Western blotting, immunofluorescence, Masson’s trichrome Sirius red staining. In addition, normal hepatocyte MIHA hepatic stellate LX-2 used as vitro models experimentally validate effects immunological, proteomic metabolomic technologies. Results We established relatively short term murine model found mainly accumulated liver. scRNA-seq performed TCS-treated control group profiled expressions > 76,000 cells belonging 13 major types. Among these types, hepatocytes (HSCs) significantly increased group. that promoted fibrosis-associated proliferation hepatocytes, which Gata2 Mef2c are driving TFs. Background: Previous studies aiming at relating exposure to phenols and phthalates with child social behavior characterized using one or a few spot urine samples, resulting in substantial misclassification. Moreover, early infancy was rarely studied. Objectives: We aimed examine the associations of cohort improved assessment priori identify chemicals supported by higher weight evidence. Methods: Among 406 mother–child pairs from French Assessment Air Pollution during Pregnancy Effect on Health (SEPAGES) cohort, 25 phenols/phthalate metabolites were measured within-subject pools repeated samples collected second third pregnancy trimesters (∼21 samples/trimester) 2 months 1-year age (∼7 samples/period). Social parent-reported 3 years Responsiveness Scale (SRS). A structured literature review animal human evidence performed prioritize phthalates/phenols based their likelihood affect behavior. Both adjusted linear regression Bayesian Weighted Quantile Sum (BWQS) models fitted. False discovery rate (FDR) correction applied only nonprioritized chemicals. Results: Prioritized compounds included bisphenol A, S, triclosan (TCS), diethyl-hexyl phthalate (ΣDEHP), mono-ethyl (MEP), mono-n-butyl (MnBP), mono-benzyl (MBzP). With exception bisphenols, which showed mixed pattern positive negative pregnant mothers neonates, prenatal observed. Most observed prioritized 1-y-old infants: Each doubling urinary TCS (β=0.78; 95% CI: 0.00, 1.55) MEP (β=0.92; −0.11, 1.96) concentrations associated worse total SRS scores, whereas MnBP ΣDEHP Awareness (β=0.25; 0.01, 0.50) Communication (β=0.43; −0.02, 0.89) respectively. BWQS also suggested [Beta 1=1.38; credible interval (CrI): −0.18, 2.97], (Beta 1=0.37; CrI: 0.06, 0.70), 1=0.91; 0.31, 1.53) scores per quartile increase mixture assessed infants. The did not remain after FDR correction, benzophenone-3 infants, suggestively (corrected p=0.07). Discussion: search allowed us adapt our statistical analysis according create corpus experimental epidemiological knowledge better interpret findings. Perfluorooctane sulfonate (PFOS), a new type of persistent organic pollutant in the environment water, has drawn significant attention recent years due to its widespread prevalence and high toxicity. Neurotoxicity is regarded as one major toxic effects PFOS, while research studies on PFOS-induced depression underlying mechanisms remain scarce. In this study, behavioral tests revealed depressive-like behaviors PFOS-exposed male mice. Neuron damages including pyknosis staining deepening were identified through hematoxylin eosin staining. Then, we noticed elevation glutamate proline levels well decline glutamine tryptophan levels. Proteomics analysis 105 differentially expressed proteins that change dose-dependent manner PFOS exposure activated glutamatergic synapse signaling pathway, which further confirmed by Western blot, data consistent with findings proteomics analysis. Additionally, downstream cyclic AMP-responsive element-binding protein (CREB)/brain-derived neurotrophic factor (BDNF) synaptic plasticity-related postsynaptic density 95, synaptophysin, downregulated. Our results highlight may inhibit plasticity hippocampus via CREB/BDNF pathway cause
