TROP-2, a tumor-associated antigen, has been implicated in the progression of various epithelial tumors. Due to its favorable expression profile, TROP-2 emerged as promising target for antibody–drug conjugates (ADCs) based anti-tumor therapies. Although ADCs have shown efficacy cancer treatment, their application solid tumors is hindered by high molecular weight, poor tumor penetration, and release cytotoxic molecules. Therefore, recombinant immunotoxin was developed on shark-derived variable domain immunoglobulin new antigen receptor (VNAR) antibody. VNARs are only one-tenth size IgG antibodies possess remarkable tissue penetration capabilities stability. In this study, shark VNAR phage display library created, leading identification VNAR-5G8 that targets TROP-2. exhibited affinity cellular internalization ability towards cells expressing levels Epitope analysis revealed recognizes hidden epitope consisting CRD TY-1 Subsequently, fused with truncated form Pseudomonas exotoxin (PE38) create (5G8-PE38), which significant activity vitro vivo. Overall, study highlights promise 5G8-PE38 valuable candidate therapy.

Food safety remains a significant global challenge that affects human health. Various hazards, including microbiological and chemical threats, can compromise food throughout the supply chain. To address issues ensure public health, it is necessary to adopt rapid, accurate, highly specific detection methods. Immunoassays are considered be an effective method for of sensitive biochemical indicators provide efficient platform identification hazards. In immunoassays, antibodies function as primary recognition elements. Nanobodies have potential valuable biomolecules in diagnostic applications. Their distinctive physicochemical structural characteristics make them excellent candidates development reliable assays, promising alternatives monoclonal polyclonal antibodies. Herein, we summarize comprehensive overview status prospects nanobody-based immunoassays ensuring safety. First, begin with historical perspective on nanobodies their unique characteristics. Subsequently, explore definitions boundaries immunosensors, before discussing applications testing emerged over past five years, follow different highlighting advantages traditional Finally, directions challenges discussed. Due remarkable sensitivity, specificity versatility, hold great promise revolutionizing health well-being.

IgE-mediated allergies represent a major health problem in the modern world. Apart from allergen-specific immunotherapy (AIT), only disease-modifying treatment, researchers focus on biologics that target different key molecules such as allergens, IgE, or type 2 cytokines to ameliorate allergic symptoms. Single-domain antibodies, nanobodies, are newcomers biotherapeutics, and their huge potential is being investigated various research fields since discovery 30 years ago. While they dominantly applied for theranostics of cancer treatment infectious diseases, nanobodies have become increasingly substantial allergology over last decade. In this review, we discuss prerequisites consider be important generating useful nanobody-based drug candidates treating allergies. We further summarize available data used allergen monitoring detection probes therapeutic approaches. reflect limitations addressed during development process, vivo half-life immunogenicity. Finally, speculate about novel application formats allergy might future.

Bacteriophages are emerging as a promising alternative in combating antibiotic-resistant bacteria amidst the escalating global antimicrobial resistance crisis. Recently, there has been notable resurgence of interest phages, prompting extensive research into their therapeutic potential. Beyond conventional microbiology and virology techniques, such genomics proteomics, novel phenotypic chemical characterization methods being explored. Among these, is growing vibrational spectroscopy, especially advanced modalities surface-enhanced Raman spectroscopy (SERS), tip-enhanced (TERS), atomic force microscopy-infrared (AFM-IR), which offer improved sensitivity spatial resolution. This review explores spectrum uses for bacteriophages, including its role diagnostics, biosensing, phage detection, assistance phage-based therapy, advancing basic research.
