Abstract Vascular calcification (VC) in type 2 diabetes (T2D) poses a serious threat to the life and health of patients. However, its pathogenesis remains unclear, resulting lack effective treatment for root cause. It is found that both intestinal Bacteroides fragilis (BF) peripheral M2 monocytes/macrophages are significantly elevated patients with T2D VC. macrophages identified as significant risk factor Both BF their extracellular vesicles (EV) promote VC facilitate macrophage polarization. Macrophages clearance antagonized EV‐induced mice. Mechanistically, EV‐rich double‐stranded DNA (dsDNA) activates stimulator interferon response cGAMP interactor 1 (Sting), promotes myocyte enhancer 2D (Mef2d) phosphorylation, upregulates tribbles pseudokinase (Trib1) expression, induces Concurrently, Mef2d activated by EV targets expression pro‐calcification Serpine1, thereby exacerbating Clinical studies have shown Serpine1 blood closely associated In summary, this study reveals Trib1 through EV‐Sting‐Mef2d pathway induce polarization serpin family E member (Serpine1) aggravating The findings provide new theoretical experimental bases optimizing strategies prevention

As cell-free nanotherapeutics, extracellular vesicles derived from mesenchymal stem cells (MSC-EVs) have shown potential therapeutic action against liver diseases. However, their effects on autoimmune hepatitis (AIH) are not yet well understood. In this study, we utilized a well-established concanavalin A (Con A)-induced fulminant mouse model to investigate the of MSC-EVs AIH. We found that provide significant protection Con A-induced in C57BL/6 male mice, with effectiveness being critically dependent gut microbiota. modulate composition microbiota, particularly by increasing abundance norank_f__Muribaculaceae, and impact metabolic profiles, leading amelioration injury. The identification Acetyl-DL-Valine as protective metabolite underscores targeting gut‒liver axis interactions Overall, our data demonstrate exhibit nanotherapeutic new insights into treatment hepatitis.

Hypertension, commonly known as high blood pressure, is a significant health issue that increases the risk of cardiovascular diseases, stroke, and renal failure. This condition broadly encompasses both primary secondary forms. Despite extensive research, underlying mechanisms systemic arterial hypertension-particularly hypertension, which has no identifiable cause affected by genetic lifestyle agents-remain complex not fully understood. Recent studies indicate an imbalance in gut microbiota, referred to dysbiosis, may promote affecting pressure regulation through metabolites such short-chain fatty acids trimethylamine N-oxide. Current antihypertensive medications face limitations, including resistance adherence issues, highlighting need for novel therapeutic approaches. Stem cell therapy, emerging field regenerative medicine, shows promise addressing these challenges. cells, with mesenchymal stem cells being prime example, have regenerative, anti-inflammatory, immunomodulatory properties. Emerging research indicates can modulate reduce inflammation, improve vascular health, potentially aiding management. Research shown positive impact on microbiota various disorders, suggesting their potential role treating hypertension. review synthesizes recent interactions between By offering thorough analysis current literature, it highlights key insights, uncovers critical gaps, identifies trends will inform guide future investigations this rapidly advancing field.

Nonalcoholic fatty liver disease (NAFLD) represents an increasing public health concern. The underlying pathophysiological mechanisms of NAFLD remains unclear, and as a result, there is currently no specific therapy for this condition. However, recent studies focus on extracellular vesicles (EVs) novelty in their role cellular communication. An imbalance the gut microbiota composition may contribute to progression NAFLD, making gut-liver axis promising target therapeutic strategies. This review aims provide comprehensive overview EVs NAFLD. Additionally, exosome-like nanovesicles derived from plants (PELNs) probiotics-derived (postbiotics) have demonstrated potential re-establish intestinal equilibrium modulate microbiota, thus offering alleviate via axis. Betaine has been proposed as a low-cost source of methyl groups in poultry feed, replacing methionine and choline. The present study aimed to investigate the effect betaine on growth performance, metabolism, transfer broilers aged 1 21 days fed low-methionine diet. A total 960 one-day-old male were randomly divided into four groups: positive control (0.62% diet), negative (0.37% two treatment diet plus either 1500 or 3000 mg betaine/kg diet). Chicks had highest feed-to-gain ratio (P < 0.05), but no significant difference final body weight, average daily gain, feed intake, mortality. Serum S-adenosyl-L-methionine homocysteine higher at diet; whereas serum S-adenosylhomocysteine exhibited opposite trend. Except for DNA methyltransferase 1, key enzymes metabolites involved hepatic single-carbon pathway showed levels declined thereafter. Furthermore, promoted dose-dependent mRNA protein expression metabolic cycle transferase pathways chicks methionine-deficient diets. In conclusion, while addition did not significantly improve performance 1-21 days, inclusion effectively stabilized metabolism
