Stachydrine, also known as proline betaine, is a prominent constituent of traditional Chinese herb Leonurus japonicus , renowned for its significant pharmacological effects. Chinese motherwort (Leonurus japonicus), a member of Lamiaceae family, is commonly used medicinal herb for treating obstetrical and gynecological diseases, producing over 280 officinal natural products. Due to limited genomic resources, little progress has been made in deciphering the biosynthetic pathway valuable products L. japonicus. Here, we de novo assembled japonicus genome using high-coverage ONT long reads Hi-C reads. The chromosome-level assembly contained ten chromosomes representing 99.29% 489.34 Mb sequence with contig scaffold N50 7.27 50.86 Mb, respectively. Genome validations revealed BUSCO LAI score 99.2% 21.99, respectively, suggesting high quality assembly. Using transcriptomic data from various tissues, 22,531 protein-coding genes were annotated. This study investigates the genetic diversity and evolutionary relationships within genus Leonurus , focusing on species found in Uzbekistan. Samples collected from Tashkent region sequenced, additional ITS data 21 representatives of Lamiaceae family retrieved GenBank. Basil, Ocimum basilicum L., is a widely cultivated aromatic herb, prized for its culinary and medicinal uses, predominantly owing to unique aroma, primarily determined by eugenol Genovese cultivars or methyl chavicol Thai cultivars. To date, comprehensive basil reference genome has been lacking, with only fragmented draft available. fill this gap, we employed PacBio HiFi Hi-C sequencing construct homeolog-phased chromosome-level basil. The tetraploid was assembled into 26 pseudomolecules further categorized subgenomes. High levels of synteny were observed between the two subgenomes but comparisons Salvia rosmarinus show collinearity quickly breaks down in near relatives.
