Abstract Herein, binding affinities of one‐walled meso ‐phenylboronic acid functionalized calix[4]pyrroles (C4Ps, 3 a & b ) with diverse anions including spherical, linear, trigonal, and tetrahedral through UV‐Vis spectroscopy have successfully been revealed in acetonitrile solvent. The C4P derivative was newly synthesized from an easily accessible 3‐acetylphenylboronic via both conventional as well green protocols. Calix[4]pyrroles are an important class of oligopyrrolic macrocycles and have found applications in many diverse fields including anion recognition. To modulate the properties calix[4]pyrrole, several structural modifications realized. The core-expansion has attracted extra attention as it provides larger cavity-size compared to parent calix[4]pyrrole(s). This review highlights synthetic development various core-expanded calix[4]pyrroles their anion-binding properties. Emphasis is given changes binding observed with expanded versions calix[4]pyrrole(s) both solution solid states. calix[4]pyrrole do not always show higher affinities for anions anticipated. Rather, they display reduced anions. truncated form or asymmetric nature does probably allow access all available sites hence observed. receptors which contain a greater number somehow rigid preorganized apparently enhanced relative constants halide series indicate that enlarged molecules more beneficial largest iodide among others. However, most selectivity towards smallest fluoride over other studied.

The ongoing challenge of multidrug-resistant Mycobacterium tuberculosis underscores the urgent need for new therapeutic agents that address both (TB) and associated oxidative stress. This study explores synthesis bioactivity six novel calix[4]pyrrole derivatives fused with amino acids, using an optimized HATU DIPEA-mediated coupling method to enhance yield stability. These were subsequently assessed their antitubercular antioxidant activities. By incorporating particularly aromatic ones, into framework, aims investigate potential as dual-function candidates managing TB stress-related complications.
