Learning and memory disorders decreased neuroplasticity are the main clinical manifestations of age-induced cognitive dysfunction. Orexin A (OxA) has been reported to show abnormally elevated expression in cerebrospinal fluid (CSF) patients with Alzheimer's disease (AD) be associated impairment. Here, we further assessed whether excitatory neurotransmitter OxA is involved function senescence-accelerated mouse prone 8 (SAMP8) mice. In this study, investigated mechanism by using behavioral tests, CSF microdialysis, immunofluorescence, toluidine blue staining, gene silencing, transmission electron microscopy, Western blotting. The results showed that 10 Hz electroacupuncture (EA) effectively alleviated learning impairment 7-month-old SAMP8 mice, reduced levels CSF, increased level glutamate, pathological damage hippocampal tissue, improved synaptic structure, enhanced transmission, regulated cAMP/PKA/CREB signaling pathway-related proteins. These suggest EA enhances mice regulating OxA-mediated pathway, thus improving function. findings may beneficial for prevention treatment
