Abstract Threat and extinction memories are crucial for organisms’ survival in changing environments. These believed to be encoded by separate ensembles of neurons the brain, but their whereabouts remain elusive. Using an auditory fear-conditioning paradigm male mice, here we discovered that two distinct projection neuron subpopulations physical proximity within insular cortex (IC), targeting central amygdala (CeA) nucleus accumbens (NAc), respectively, encode fear memories. Reciprocal intracortical inhibition these IC gates emergence either or memory. rabies-virus-assisted tracing, found IC-NAc preferentially innervated intercortical inputs from orbitofrontal (OFC), specifically enhancing override results demonstrate serves as operation node harboring decipher memory under top-down executive control OFC.

Abstract Traumatic brain injury (TBI) is a risk factor for posttraumatic stress disorder (PTSD). Augmented fear defining characteristic of PTSD, and the amygdala considered main region to process fear. The mechanism by which involved in conditioning after TBI still unclear. Using single‐nucleus RNA sequencing (snRNA‐seq), transcriptional changes cells are investigated. In total, 72 328 nuclei obtained from sham groups. 7 cell types, analysis differentially expressed genes (DEGs) reveals widespread each type identified. vivo experiments, it demonstrated that Decorin (Dcn) expression excitatory neurons significantly increased TBI, Dcn knockout mitigates TBI‐associated conditioning. Of note, this effect caused Dcn‐mediated decrease perineuronal nets (PNNs), affect glutamate‐ γ ‐aminobutyric acid balance amygdala. Finally, results suggest functions interacting with collagen VI α 3 (Col6a3). Consequently, findings reveal different types provide direct evidence relieves regulating PNNs.

Animals use prior experience to assign absolute (good or bad) and relative (better worse) value new experience. These learned values guide appropriate later decision making. Even though our understanding of how the valuation system computes is relatively advanced, mechanistic underpinnings are unclear. Here, we uncover mechanisms aversive in Drosophila. Three types punishment-sensitive dopaminergic neurons (DANs) respond differently electric shock intensity. During learning, these DANs drive intensity-scaled plasticity at their respective mushroom body output neuron (MBON) connections code value. In contrast, by comparing current previous experiences, MBON-DAN network can using specific recruiting a subtype reward-coding DANs. Behavioral physiological experiments revealed that DAN assigns "better than" lesser two experiences. This study therefore highlights appetitive-aversive interactions within MB compare sequential experiences learn

Extinction memory retrieval is influenced by spatial contextual information that determines responding to conditioned stimuli (CS). However, it poorly understood whether representations are imbued with emotional values support selection. Here, we performed activity-dependent engram tagging and in vivo single-unit electrophysiological recordings from the ventral hippocampus (vH) while optogenetically manipulating basolateral amygdala (BLA) inputs during formation of cued fear extinction memory. During when CS acquire safety properties, found CS-related activity vH reactivated sleep consolidation was strengthened upon retrieval. Moreover, facilitated context exhibited precise coding its affective zones. Last, these patterns along were dependent on glutamatergic transmission BLA learning. Thus, relies stimulus instructed BLA.
