The rapid and coordinated propagation of neural activity across the brain provides foundation for complex behavior cognition. Technical advances neuroscience subfields have advanced understanding these dynamics, but points convergence are often obscured by semantic differences, creating silos subfield-specific findings. In this review we describe how a parsimonious conceptualization state as fundamental building block whole-brain offers common framework to relate findings scales species. We present examples diverse techniques commonly used study states associated with physiology higher-order cognitive processes, discuss integration them will enable more comprehensive mechanistic characterization dynamics that crucial survival disrupted in disease.

Abstract Reinforcement feedback can improve motor learning, but the underlying brain mechanisms remain underexplored. In particular, causal contribution of specific patterns oscillatory activity within human striatum is unknown. To address this question, we exploited a recently developed non-invasive deep stimulation technique called transcranial temporal interference (tTIS) during reinforcement learning with concurrent neuroimaging, in randomized, sham-controlled, double-blind study. Striatal tTIS applied at 80 Hz, not 20 abolished benefits on learning. This effect was related to selective modulation neural striatum. Moreover, increased neuromodulatory influence frontal areas involved These results show that non-invasively and selectively modulate striatal mechanism expanding our tools for study relationships between structures behaviour.

Over the past decade, research has shown that primary motor cortex (M1), brain's main output for movement, also responds to rewards. Abstract Accurate senses depend on high-fidelity encoding by sensory receptors and error-free processing in the brain. Progress has been made towards restoring damaged receptors. However, methods for on-demand treatment of impaired central are scarce. Prior invasive studies demonstrated that continuous vagus nerve stimulation (VNS) rodents can activate locus coeruleus-norepinephrine system to rapidly improve processing. Here, we investigated whether transcutaneous VNS improves performance humans. We conducted three sham-controlled experiments, each with 12 neurotypical adults, measured effects metrics auditory visual performance, heart rate variability (HRV). Continuous was delivered cervical (tcVNS) or auricular (taVNS) branches while participants performed psychophysics tasks passively viewed a display. Relative sham stimulation, tcVNS improved 37% ( p = 0.00052) 23% 0.038). Participants lower during conditions experienced larger tcVNS-evoked improvements 0.0040). Lastly, increased HRV passive viewing, corroborating vagal engagement. No evidence an effect taVNS observed. These findings validate effectiveness humans position it as method interventions impairments associated dysfunction.

Abstract Inflammation, caused by accumulation of inflammatory cytokines from immunocytes, is prevalent in a variety diseases. Electro-stimulation emerges as promising candidate for inhibition. Although electroacupuncture free surgical injury, it faces the challenges imprecise pathways/current spikes, and insufficiently defined mechanisms, while non-optimal pathway or spike would require high current amplitude, which makes electro-stimulation usually accompanied damage complications. Here, we propose neuromorphic based on atomically thin semiconductor floating-gate memory interdigital circuit. Direct stimulation achieved wrapping sympathetic chain with flexible electrodes are programmable to fire bionic thus minimizing nerve damage. A substantial decrease (73.5%) cytokine IL-6 occurred, also enabled better efficacy than commercial stimulator at record-low currents damage-free neurons. Additionally, using transgenic mice, anti-inflammation effect determined β2 adrenergic signaling myeloid cell lineage (monocytes/macrophages granulocytes).

Major depressive disorder (MDD) is often marked by impaired motivation and reward processing, known as anhedonia. Many patients do not respond to first-line treatments, improvements in can be slow, creating an urgent need for rapid interventions. Recently, we demonstrated that transcutaneous auricular vagus nerve stimulation (taVNS) acutely boosts effort invigoration healthy participants, but its effects on depression remain unclear.
