Sex plays a crucial role in human brain development, aging, and the manifestation of psychiatric neurological disorders. However, our understanding sex differences functional organization their behavioral consequences has been hindered by inconsistent findings lack replication. Here, we address these challenges using spatiotemporal deep neural network (stDNN) model to uncover latent dynamics that distinguish male female brains. Our stDNN accurately differentiated brains, demonstrating consistently high cross-validation accuracy (>90%), replicability, generalizability across multisession data from same individuals three independent cohorts (N ~ 1,500 young adults aged 20 35). Explainable AI (XAI) analysis revealed features associated with default mode network, striatum, limbic exhibited significant (effect sizes > 1.5) sessions cohorts. Furthermore, XAI-derived predicted sex-specific cognitive profiles, finding was also independently replicated. results demonstrate are not only highly replicable generalizable but behaviorally relevant, challenging notion continuum male-female organization. underscore as biological determinant organization, have implications for developing personalized biomarkers disorders, provide innovative AI-based computational tools future research.

Frontotemporal dementia (FTD) encompasses a group of heterogeneous neurodegenerative disorders. Aside from genetic cases, its diagnosis is challenging, particularly in the early stages when symptoms are ambiguous, and structural neuroimaging does not reveal characteristic patterns. The authors performed comprehensive literature search through MEDLINE, Scopus, Web Science databases to gather evidence aid diagnostic process for suspected FTD patients, phases, even sporadic ranging established promising tools. Blood-based biomarkers might help identify very neuropathological guide further evaluations. Subsequently, neurophysiological measures reflecting functional changes cortical excitatory/inhibitory circuits, along with assessing brain network, connectivity, metabolism, perfusion alterations, could detect specific associated decades before symptom onset. As advances, cognitive-behavioral profiles atrophy patterns emerge, distinguishing subtypes. Emerging disease-modifying therapies require patient enrollment. Therefore, paradigm shift needed - relying on typical cognitive advanced cases widely applicable biomarkers, primarily fluid and, subsequently, where appropriate. Additionally, exploring subjective complaints behavioral detected by home-based technologies be crucial diagnosis.

Abstract Altered functional connectivity precedes structural brain changes and symptoms in dementia. Alzheimer’s disease is the largest contributor to dementia at population level, disrupts brain’s default-mode network (DMN). We investigated whether a neurobiological model of DMN effective could predict future diagnosis single-participant level. applied spectral dynamic causal modeling resting-state magnetic resonance imaging data nested case–control group from UK Biobank, including 81 undiagnosed individuals who developed up nine years after imaging, 1,030 matched controls. Dysconnectivity predicted both incidence (AUC = 0.82) time ( R 0.53), outperforming models based on structure connectivity. also evaluated associations between dysconnectivity major risk factors for dementia, revealing strong relationships with polygenic social isolation. Neurobiological may facilitate early detection supporting rational deployment targeted dementia-prevention strategies.

Abstract The default mode network (DMN) is a widely distributed, intrinsic brain thought to play crucial role in internally directed cognition. present study employs stereo-EEG 13 human patients, obtaining high resolution neural recordings across multiple canonical DMN regions during two processes that have been associated with creative thinking: spontaneous and divergent thought. We probe these DMN-associated higher cognitive functions through mind wandering alternate uses tasks, respectively.
