Abstract
Purpose
The
mechanisms
underlying
reversible
unconsciousness
induced
by
general
anesthetics
remain
unclear.
This
study
aimed
to
investigate
the
effects
of
four
commonly
used
on
neuronal
spiking
patterns
in
layer
5
medial
prefrontal
cortex
(mPFC).
Methods
In
vivo
multi-channel
recordings
were
performed
mPFC
a
mouse
model.
Neuronal
regular-spiking
and
fast-spiking
neurons
measured
compared
across
wakefulness
loss
righting
reflex
(LORR),
recovery
(RORR).
Four
anesthetic/sedative
drugs
(sevoflurane,
propofol,
ketamine,
dexmedetomidine)
tested.
Results
During
LORR,
most
cortical
inhibited,
while
small
subset
was
excited.
Fast-spiking
exhibited
significant
suppression
all
anesthetics.
Among
these,
firing
rate
inhibited
closely
associated
with
transitions
between
LORR
RORR.
Sevoflurane,
dexmedetomidine
similar
modulatory
neurons,
whereas
ketamine
stronger
excitatory
both
regular-
neurons.
Conclusions
exert
comparable
mPFC,
induces
distinct
effects.

Background
Increasing
evidence
indicates
that
transcutaneous
auricular
vagus
nerve
stimulation
(taVNS)
can
promote
the
recovery
of
consciousness
levels
in
patients
with
prolonged
disorders
(pDOC).
However,
previous
clinical
studies
have
shown
inconsistent
efficacy
taVNS
treating
pDOC.

could
be
because
no
research
has
clearly
defined
indications
for
treatment

Abstract
Perceptual
multistability
has
been
studied
for
centuries
using
a
diverse
collection
of
approaches.
Insights
derived
from
this
phenomenon
range
core
principles

Abstract
The
role
of
the
prefrontal
cortex
(PFC)
in
consciousness
is
hotly
debated.
Frontal
theories
argue
that
PFC
necessary
for
consciousness,
while
sensory
propose
arises
from
recurrent
activity
posterior
alone,
with
resulting
mere
act
reporting.
To
resolve
this
dispute,
we
re-analysed
an
EEG
dataset
30
participants
a
no-report
inattentional
blindness
paradigm
where
faces
are
(un)consciously
perceived.
Dynamic
causal
modelling
was
used
to
estimate
effective
connectivity
between
key
contended
brain
regions,
and
cortices.
Then,
second-level
parametric
empirical
Bayesian
model
conducted
determine
how
modulated
by
awareness
task-relevance.
While
initial
data-driven
search
could
not
corroborate
neither
nor
frontal
more
directed
hypothesis-driven
analysis
revealed
strong
evidence
